维拉帕米对肝星状细胞凋亡和Bax表达的影响

摘要 目的:观察维拉帕米对人肝星状细胞(Hepatic stellate cells,HSCs)凋亡以及凋亡相关蛋白Bax表达的影响。方法:采用不同浓度的维拉帕米对HSCs处理48h,CCK-8试剂盒(Cell Counting Kit-8)检测细胞增殖抑制率,HE染色观察细胞形态超微结构的变化,流式细胞仪AnnexinV/PI双染法检测HSCs细胞的凋亡率,免疫组化SABC法检测Bax表达的变化。结果:维拉帕米可抑制HSCs的增殖,随浓度的增加,抑制作用增强,其IC50值为0.15±0.01mol.L-1;HE染色可见维拉帕米组出现不同程度的细胞体积变小、核浓缩深染等凋亡形态改变;AnnexinV/PI双染法检测显示,不同浓度的维拉帕米均可诱导HSCs细胞凋亡,与阴性对照组比较,差异有统计学意义(P<0.05);免疫组化检测显示,与对照组比较,维拉帕米组Bax表达增强(P<0.05)。结论:维拉帕米可诱导HSCs凋亡,增强Bax的表达。结果:维拉帕米;肝星状细胞;凋亡; Objective: To observe the effects of verapamil on apoptosis and bax expression of human HSCs. Methods: HSCs were incubated with different concentration of verapamil for 48 h, cell proliferation was assessed by cell counting kit-8, morphologic change was detected by HE staining, the rate of apoptosis was tested by SABC of irnmunohistochemical method. Results. Verapamil can inhibit the proliferation of HSCs in a dose-dependent manner, the IC50 value as 0. 155:0. 01 mol·L^-1. HE staining showed different changes of cell apoptosis such as soma shrinking, nucleus concentrating and anachromasis. The apoptosis of HSCs was in-duced by different concentration of verapamil, which was different from the negative control group (P〈0. 05). The expression of bax of HSCs cells after treatment of verapamil was increased, which was different from the negative control group (P〈0. 05). Conclu-sion: Verapamil can induce the apoptosis and increase the expression of Bax of HSCs.
