通脑丸干预脑动脉硬化症慢性脑缺血损伤的作用机制研究 被引量:3

摘要 目的:探讨通脑丸干预脑动脉硬化症慢性脑缺血损伤的作用机制。方法:以双肾双夹法复制肾性高血压进而建立脑动脉硬化症模型大鼠为研究对象,持续灌胃14周后,观察各组大鼠血脂、血清学相关指标、脑组织血管内皮生长因子(VEGF)含量的变化。结果:治疗第14周末时,与模型组比较,步长脑心通组,通脑丸高、低剂量组总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白胆固醇(LDL-C)水平、血清丙二醛(MDA)含量均有显著降低,高密度脂蛋白胆固醇(HDL-C)、血清超氧化物歧化酶(SOD)含量、谷胱甘肽过氧化物酶(GSH-PX)活性、VEGF的表达显著升高(P<0.05)。与步长脑心通组比较,通脑丸高、低剂量组的TC、TG、LDL-C三项血脂指标降低显著,HDL-C、VEGF的表达显著升高;通脑丸高、低剂量组SOD含量、MDA含量、GSH-PX活性无统计学意义(P>0.05)。通脑丸高剂量组TC、TG、LDL-C、VEGF优于低剂量组,差异有显著性意义(P<0.05)。结论:通脑丸可通过提高SOD、GSH-PX活性活力,上调脑动脉硬化症模型大鼠脑组织VEGF表达,降低MDA的含量,从而减轻动脉粥样硬化发展过程中的自由基损伤,发挥抗脑细胞损伤的作用,其效应与步长脑心通相似。 Objective: To explore the mechanism of TongNaoWan intervening the injury of chronic cerebral is- chemia induced by cerebral arteriosclerosis. Method: Rat models were established through two-kidney two-clip method replicating renal hypertension. Changes of the related indexes of blood fat, serology and the content of VEGF in cerebral tissue of rats were observed in different groups after lavage for 14 consecutive weeks. Result: By the end of 14th week, compared with model group, the contents of TC, TG, LDL-C and MDA in the groups of BuChang NaoXinTong, high and low dosage of TongNaoWan were decreased signficantly, the contents of HDL-C and SOD, the activity of GSH-PX and the expression of VEGF were remarkably improved (P〈0.05). Compared with BuChang NaoXinTong group, the decrease of TC, TG and LDL-C was greater in the high and low dosage groups of TongNaoWan, the expressions of HDL-C and VEGF were raised significantly; it showed no statistical meaning in the contents of SOD, MDA and the activity of GSH-PX of the high and low dosage groups of Tong- NaoWan (P〉0.05). The contents ofTC, TG, LDL-C and VEGF in the high dose group of TongNaoWan were superi- or to low dosage group among the groups of TongNaoWon, the difference was significant (P〈0.05). Conclusion: TongNaoWan could raise the activity of SOD and GSH-PX, improve the expression of VEGF in cerebral tissue of rat with cerebral arteriosclerosis and lower the content of MDA, therefore to relieve the damage of free radical during the development of cerebral arteriosclerosis and play the role of anti-injury cerebral cells, the function is similar to BuChang NaoXinTong.
