摘要
目的:探讨淫羊藿苷和黄芪甲苷对皮质酮诱导的原代培养海马神经元损伤的保护作用。方法:体外培养新生大鼠海马神经元,加入不同浓度的皮质酮(0.01-5μmol/L)诱导原代培养海马神经元建立皮质酮损伤模型。培养体系中加入不同浓度的淫羊藿干和黄芪甲苷预处理。采用MTT法和LDH释放法检测对神经元存活率的影响;倒置显微镜下观察神经元的生长发育形态;采用Caspase-3分光光度法检测神经元凋亡。结果:与皮质酮组比较,中高剂量淫羊藿苷可明显增加海马神经元活力,降低LDH释放,具有显著性差异(P<0.05,P<0.01)。淫羊藿苷干预后,核固缩的神经元比例大幅降低。Caspase-3活性显著性降低(P<0.01),而黄芪甲苷组对神经元无明显保护作用。结论:补肾组分淫羊藿苷对皮质酮诱导的原代培养海马神经元损伤具有保护作用,其可能与抑制神经元凋亡有关。而益气组分黄芪甲苷无明显保护作用。
Objective: To investigate the neuroprotective effects of icariin and astragaloside Ⅳon corticosterone-induced apoptosis in primary cultured rat hippocampal neurons. Methods: Neonatal rat hippocampal neurons were cultured in vitro with different concentrations of corticosterone (0.01-5μmol/L) induced primary cultured hippocampal neurons to establish neuronal damage model. The culture system was added different concentrations of the icariin and astragaloside Ⅳ pretreatment. Neuronal viability was detected by MTT assay and lactate dehydrogenase releasing methods; The growth and development of the neuron morphology was observered with an inverted microscope; Neuronal apoptosis was determined by caspase-3 spectrophotometric detection. Result: Compared with the model group, medium and high dosage of icariin significantly increased the hippocampal neuronal viability, and decreased the release of LDH, and there were significant differences(P〈0.05, P〈0.01). After intervention by icariin, the karyopyknosis neuronal proportion significantly decreased. The caspase-3 viability significantly decreased(P〈0.01). Compared with the model group, astragaloside group didn't have significant protective effect. Conclusion: The current results demonstrated that icariin could suppress corticosterone-induced primary cultured rat hippocampal neuron apoptosis, which may be related to the inhibition of neuronal apoptosis. But astragaloside Ⅳ didn't have significant protective effect.
