摘要
目的探讨山药多糖预处理对肾缺血再灌注损伤大鼠肾脏缺氧诱导因子-1α(HIF-1α)和血红素加氧酶-1(HO-1)表达的影响。方法将健康雄性SD大鼠30只,随机分为假手术组、模型组、山药多糖预处理组,制备大鼠肾缺血再灌注损伤模型。术前1周,山药多糖预处理组每日予以山药多糖(200 mg.kg-1)灌胃,假手术组和模型组每日予以生理盐水(10 mL.kg-1)灌胃,再灌注6 h后检测各组大鼠血尿素氮(BUN)和血清肌酐(Scr)的含量,Western blot和RT-PCR法分别检测各组肾组织HIF-1α蛋白及mRNA、HO-1蛋白的表达水平。结果山药多糖预处理组BUN、Scr含量明显低于模型组(P<0.01),肾组织HIF-1αmRNA及蛋白、HO-1蛋白的表达明显高于模型组(P<0.01)。结论山药多糖预处理对肾缺血再灌注损伤具有明显保护作用,其机制可能为通过上调肾组织中HIF-1α的表达进而诱导HO-1的表达。
Objective To explore the impacts of Chinese yam polysaccharide preconditioning (CYPP) on the expressions of HIF -1α and HO - 1 in the rats of renal ischemic reperfusion injury. Methods Thirty SD healthy male rats were randomized into three groups, a pseudo -operation group, a model group and a CYPP group. The rat model of renal ischemic reperfusion injury was prepared. In the CYPP group, the gastric perfusion of Chinese yam polysaccharide(200 mg · kg-1)was applied one week before operation. In the pseudo- operation group and the model group, the physical saline (10 mL · kg-1) was used for gastric perfusion every day. 6h after reperfusion,the contents of blood BUN and Scr were detected in the rats of each group. Western blot and RT - PCR methods were adopted to detect the expressions of HIF -1α, mRNA and HO - 1 protein of renal tissue in each group. Results In the CYPP group, BUN and Scr contents were lower apparently than those in the model group( P 〈 0.01 ). The expressions of HIF -1α, mRNA and HO - 1 pro- tein of renal tissue were higher apparently than those in the model group (P 〈 0.01 ). Conclusion Chinese yam polysaccharide preconditioning plays the apparent protective role in renal ischemic reperfusion injury. The mechanism of it is possibly relevant with the up - regulation of HIF -1α in renal tissue to induce HO - 1 expression.
