摘要
目的探讨有氧运动联合丝瓜络对糖尿病大鼠的治疗作用及其机制。方法将40只雄性SD大鼠随机分为正常组、模型组、运动组、丝瓜络组、联合组各8只。除正常组外,另四组均采用链脲佐菌素诱导建立糖尿病模型。模型建立后2 d,运动组及丝瓜络组分别行有氧运动训练和丝瓜络10 g/kg灌胃给药,联合组同时行有氧运动和丝瓜络干预(剂量、方法同前)。干预6周后采用简易代谢笼法测定各组24 h尿量,血糖仪测定各组血糖,RT-PCR法检测肾脏髓质水通道蛋白2(AQP2)mRNA表达。结果①血糖:模型组及运动组均明显高于正常组;运动组低于模型组(P均<0.05);丝瓜络组及联合组均低于模型组,丝瓜络组与联合组比较无统计学差异。②24 h尿量:模型组、运动组、丝瓜络组、联合组均明显多于正常组;运动组、丝瓜络组及联合组均显著多于模型组(P均<0.05),联合组显著多于丝瓜络组(P<0.05)。③肾髓质AQP2 mRNA表达:模型组及运动组显著高于正常组;运动组低于模型组,但无显著差异;丝瓜络组及联合组显著低于模型组(P均<0.05);联合组显著低于丝瓜络组。结论有氧运动联合丝瓜络对糖尿病大鼠可协同发挥利尿作用,其机制可能为下调肾髓质AQP2 mRNA表达。
Objective To investigate the therapeutic effect of aerobic exercise combined with retinervus luffae fructus (RLF) on diabetic rat model and its mechanism. Methods Forty male Sprague-Dawley rats were divided into five groups randomly, including control group (group A), diabetes mellius model group (group B), exercise diabetic group (group C), retinervus luffae fructus diabetic group (group D ), retinervus luffae fructus and exercise diabetic group (group E ). Except for the control group, all diabetes mellitus rat models were induced by streptozotocin injected intraperitoneally. Two days later, the exercise diabetic group and retinervus luffae fructus diabetic group were given aerobic exercise and retinervus luffae fructus 10g/kg each day respectively. The rctinervus luffae fructus and exercise diabetic group was given both the aerobic exercise and retinervus luffae fructus 10 g/kg each day. After intervention for 6 weeks, the simple metabolic cage method was used for the determination of the rat urinary volume for 24 hours in each group. Blood glucose was determined by blood glucose meter. AQP2 mRNA expression in rat kidney was evaluated by RT-PCR. Results ( 1 ) Blood glucose : the blood glucose in group B and C was higher than that in group A, and group. C was lower than group B ( all P 〈 0.05 ) ; the blood glucose in group D and E was lower than that in group B, and no significantly statistical difference was found be- tween group D and E. (2) Urinary volume for24 hours: the urinary volume for24 hours in group 13, C, D and E was more than that in group A; the urinary volume in group C, D and E was much more than that in group B ( all P 〈 0.05 ) , and group E was much more than group D (P 〈 0.05 ). ( 3 ) AQP2 mRNA expression: the AQP2 mRNA expression in group B and C was much higher than that in group A; the AQP2 mRNA expression in group C was lower than that in group B, with no significant difference ; the AQP2 mRNA expression in group D and E was much lower than that in group B ( all P 〈0.05 ) ; the AQP2 mRNA expression in group E was much lower than that in group D. Conclusion Aerobic exercise com- bined with retinervus luffae fructus synergistically play a diuretic effect on diabetic rats, which might be related to the down- regulation of AQP2 mRNA expression in the renal medulla.
