摘要
目的探讨帕瑞昔布治疗经前期综合征(PMS)的效果及可能机制。方法将45只Wistar大鼠随机分为正常组、模型组及治疗组,每组15只。模型组及治疗组采用慢性束缚应激法制作PMS模型,持续7 d。造模第5~7天,治疗组每天肌注一次帕瑞昔布(0.72 mg/kg)。造模成功第8天处死各组大鼠。ELISA法检测外周血雌二醇(E2)、5-羟色胺(5-HT)、β-内啡肽(β-EP)水平,Western blot法检测下丘脑组织5-HT、β-EP表达。结果①与正常组比较,模型组和治疗组血浆E2水平显著升高(P均<0.05);治疗组与模型组血浆E2水平差异不显著。②与模型组比较,正常组和治疗组血浆5-HT和β-EP水平显著升高(P<0.01,P<0.05);与正常组比较,治疗组血浆5-HT和β-EP水平显著降低(P均<0.05)。③与模型组比较,正常组和治疗组下丘脑5-HT和β-EP水平显著升高(P<0.05,P<0.01);与正常组比较,治疗组下丘脑5-HT和β-EP水平显著降低(P均<0.05)。结论帕瑞昔布对PMS大鼠外周血E2水平无明显影响,但能增加5-HT和β-EP在外周血和下丘脑的表达。
Objective To investigate the effect of parecoxib treatment on premenstrual syndrome (PMS) and the pos- sible mechanism. Methods Forty-five Wistar rats were divided into 3 groups according to the results of vaginal secretion smear: normal group (group C), PMS model group (group M), and PMS model + parecoxib group( group A). Rat model of PMS was prepared by chronic restraint stress for 7 days. Group A was administered parecoxib sodium(0.72mg/kg)pre- intervention from the fifth day to the seventh day. All the rats were sacrificed on the eighth day. The estrogen 2( E2 ) ,5- hydroxytryptamie(5-HT) and β-endorphin (β-EP) levels in the peripheral blood were detected by ELISA, and the 5-HT and β-EP expression in hypothalamus was detected by Western blot. Results ( 1 ) The plasma E2 level of group M and group A were significantly higher than that in group C ( P 〈 0.05 ) ; no difference were found in group M and group A. ( 2 ) The 5-HT and β-EP levels of rat plasma in group C and group A were significantly lower than group M ( P 〈 0.01, P 〈 0.05) ; compared with group C, the 5-HT and β-EP levels of rat plasma were significantly lower in group A ( all P 〈 0.05). (3) Compared with group M, the expression of 5-HT and β-EP of hypothalamus in group C and group A were sig- nificantly higher ( P 〈 0.05, P 〈 0.01 ) and the expression'of 5-HT and β-EP of hypothalamus in group A Were significantly lower than group C ( all P 〈 0.05). Conclusion Parecoxib could not affect the E2 level of plasma , but it could increase the expression of 5-HT and β-EP in peripheral blood and hypothalamus of PMS model rats.
