HGF复合Pluronic F-127治疗大鼠下颌骨缺损的实验研究

摘要 目的:观察复合肝细胞生长因子(hepatocyte growth factor,HGF)的Pluronic F-127凝胶(下称HGF凝胶)对骨髓间充质干细胞(bone marrow mesenchymal stem cells,BMSCs)的定向募集以及对大鼠下颌骨缺损愈合的影响。方法:以Transwell小室进行HGF凝胶对BMSCs的体外趋化实验。于30只SD大鼠双侧下颌骨制造骨缺损,右侧植入HGF凝胶,左侧为空白对照,随机分为5组,每组6只,分别于术后第1、2、4、6、8周末处死,X线片观察两侧骨创愈合情况。骨钙素(bone gla protein,BGP)及CD34免疫组织化学染色后对免疫组织化学评分及新生血管数进行统计分析。结果:趋化实验显示HGF凝胶组BMSCs迁移数大于空白对照组(P<0.05)。术后X线片显示各时间点实验侧愈合较对照侧快。术后1-6周实验侧成骨较对照侧活跃(P<0.05),实验侧新生血管数大于对照侧(P<0.05);第8周两指标均无明显区别(P﹥0.05)。结论:HGF凝胶对BMSCs有趋化作用,并且有一定的促进成骨和成血管的作用。 Objective:The present study was designed to evaluate whether the compound of hepatocyte growth factor (HGF) and Pluronic F-127 gel are associated with fixed direct recruitment of bone marrow mesenchymal stem cells (BMSCs), and the healing process of bone defects. Methods:Transwell chamber was used to observe the migration of BMSCs towards HGF-gel. Bone defects were established in two sides of the mandible in 30 SD rats. In the right side of the mandible defects, HGF-gel was injected. The animals were randomly divided into 5 groups and sacrificed on the 1-, 2-, 4-, 6-, and 8-week after surgery. Both sides of the bone defects were checked by X-ray examination. The growth of new vessels and the formation of new bones in bilateral bone defects were detected by histochemical method through bone gla protein (BGP) and CD34 expression. The data of new vessels quantities and the immunoreactive score were analyzed. Results:Fixed direct recruitment of bone marrow mesenchymal stem cells (BMSCs) in HGF-gel group was more than the group of controls (P〈O.05). The bone defects of HGF-gel sides repaired faster than the control side through X-ray examination. In the healing process from the 1st week through 6th week, the formation of new bones in HGF-gel sides were more active than control sides (P〈0.05), and more new vessels were found in HGF-gel sides than the control sides (P〈0.05). In the period of 8th week, significant differences disappeared between the groups. Conclusion: HGF-gel may accelerate BMSCs migration, furthermore, it can enhance new vessels growth and new bones formation.
