摘要
目的:探讨电针治疗创伤后应激障碍(PTSD)作用机制。方法:将30只雄性SD大鼠随机均分为3组:正常组、模型组和电针治疗组(简称电针组)。后两组大鼠造模,采用国际认定的单程长时应激(SPS)方法制作PTSD模型大鼠;SPS刺激结束后,电针组大鼠给予"百会"与"足三里"低频(2 Hz)电针刺激,30min/次,每日1次,连续1周。采用逆转录聚合酶链反应(RT-PCR)方法和免疫组织化学方法分别检测3组大鼠海马神经元型一氧化氮合酶(nNOS)mRNA和蛋白的表达。结果:①RT-PCR结果显示,模型组大鼠海马nNOS mRNA表达明显高于正常组(P<0.05);电针组大鼠海马nNOS mRNA表达恢复性下调,显著低于模型组(P<0.05)。②免疫组化结果显示,模型组大鼠海马CA1区和CA3区nNOS蛋白表达明显高于正常组(P<0.05),电针组大鼠海马CA1区、CA3区nNOS蛋白表达恢复性下调,显著低于模型组(P<0.05)。各组大鼠海马CA2区nNOS蛋白表达差异无统计学意义。结论:海马nNOS表达升高可能参与PTSD的病理过程,电针下调PTSD模型大鼠海马nNOS表达可能是电针治疗PTSD的作用机制之一。
Objective To explore the mechanism of electroacupuncture (EA) in the treatment of post-traumatic stress disorder (PTSD). Methods Thirty male Sprague-Dawley rats were randomly divided into a normal group, a model group and an electroacupuncture group. The single prolonged stress (SPS) method was used to set up the PTSD models in latter two groups. After SPS Stimulation, EA group was treated with 2Hz electroacupuncture at Baihui (GV 20) and Zusanli (ST 36) for 30 min. once a day for a week. Reverse transcriptase polymerase chain reaction (RT-PCR) and immuno-histochemistry were used to detect the mRNA and protein expression of nNOS in the hippocampus of rats in the each group. Results ① The nNOS mRNA expression in hippocampus in modelgroup was higher than that in normal group (P〈0. 05). But the expression in EA group was lower significantly than that in model group (P〈0.05). ②The nNOS protein expression in hippoeampus CA1 and CA3 in model group was higher than that in normal group (P〈0, 05). But after electroaeupuneture treatment, its expression in EA group was lower significantly than that in model group (P〈0.05). The nNOS protein expression in hippoeam- pal CA2 had no differenee among all three groups, Coheirion The elevated nNOS expression in hippoeampus may he involved in the pathological proeess of PTSD. Eleetroaeupuneture play a down-regulation effects in the hip- poeampal nNOS expression, whieh may be one mechanism of eleetroaeupuneture for treatment of PTSD.
