多指标评价白芷超临界提取物-PLGA胶束制备工艺

摘要 目的筛选白芷超临界提取物-聚乳酸羟基乙酸共聚物(PLGA)载药胶束制备工艺。方法采用正交试验设计法,整体柱-HPLC测定欧前胡素、异欧前胡素含量,以载药率和包封率以及累积溶出率为指标,优化载药胶束的制备工艺。结果优选的制备工艺为PLGA与白芷超临界提取物质量比9:1,有机相与水相体积比1:20,PVA浓度2%,转速2500 r/min。结论优选的胶束制备工艺稳定,可明显提高白芷超临界提取物的溶出度,利于其体内吸收。 Objective Optimization the preparation technology of Radix Angelicae Dahuricae supercritical extraction-PLGA micelles. Methods Orthogonal design was performed to study the preparation technology of micelles. The contents of imperatorin and isoimperatorin were determined by Monolithic Silica column- high performance liquid chromatography. The encapsulation rate, drug loading efficiency and dissolution were considered as evaluation index of the micelle technology. Results The optimum preparation procedure was that the mass ratio of PLGA and Radix Angeticae Daburicae supercritical extraction was 9:1, the volume ratio of organic and water solution is 1:20, the concentration of PVA is 2%, and the stirring speed is 2500 r/min. Conclusion The optimized technology can produce stable micelles, which increases drug dissolution obviously and promotes the absorption of coumarin derivatives in body.
