摘要
采用基于LC/Q-TOF MS的代谢组学方法,对不同致病性青枯雷尔氏菌的胞外代谢物进行了分析,获得了具有显著性差异的代谢标志物,并采用PCA数据处理方法对不同致病性菌株进行分类,建立致病性的PLS-DA预测模型。结果表明,该方法可以从代谢水平上区分野生型强致病力菌株、野生型无致病力菌株以及强致病力菌株经Tn-5转座子插入导致致病力丧失的人工致弱突变菌株。对代谢标志物的差异分析结果表明,在无致病力菌株和Tn-5致弱菌株中,有12种代谢标志物与强致病力菌株具有显著性差异(P<0.05,FC≥2),并表现出相同的丰度变化趋势,预示着这些代谢标志物可能与青枯雷尔氏菌的致病性有关。
The metabolites of Ralstonia solanacearum with various pathogenic were analysed by metabonomic method based on LC/Q-TOF MS. Significant metabolic markers were used for classification pathogenic of Ralstonia solanacearurn by PCA analysis, and PLS-DA prediction model were established. The result suggest that metabonomic analysis could be used for indentify virulent Ralstonia solanacearum strain and avirulent strain and Tn-5 mutant strains. Furthermore, there were 12 metabolites showed similar trend and had significant differences between virulent strain and avirulent strains (both wide-type and Tn-5 mutant), suggesting that the 12 metabolites are probably related to the pathogincity of Ralstonia solanacearum.
