紫草中β,β'-二甲基丙烯酰阿卡宁的含量测定及不同来源药材质量评价的初步研究 被引量:5

摘要 目的:建立HPLC法测定紫草中β,β′-二甲基丙烯酰阿卡宁的含量,并对药材质量进行初步评价方法:色谱柱:Cosmosil 5C_(18)-MS-II(150 mm×4.6 mm,5μm);流动相:乙腈-水-甲酸(430:570:0.5);检测波长:275nm;流速:1.0 ml·min对8批紫草药材进行定量测定,并提取特征图谱信息,进行聚类分析和主成分分析。结果:各紫草样品中β,β′-二甲基丙烯酰阿卡宁的含量均大于3 mg·g^(-1),符合现行标准规定,但成分谱存在一定的差异。通过多变量统计分析,试验样品可分为新疆紫草、未知品种的进口紫草与滇紫草;除β,β′-二甲基丙烯酰阿卡宁外,其他成分对其质量评价也起着重要作用。结论:目前市售紫草药材的基原与质量均有较大差异,本方法准确、可靠,可用于其综合评价研究。 Objective;To establish an HPLC method for the determination ofβ,β’-dimethylacrylalkannin in Aniebiae Radix from different sources and evaluate the quality.Method:The HPLC method was developed with a Cosmosil 5C18-MS-Ⅱwaters column(150 mm×4.6 mm,5μm) with the detection wavelength of 275 nm and the column temperature of 25℃.The mobile phase was acetonitrile -water-formic aeid(430:570:0.5 ) at a flow rate of 1.0 ml·min-1.Eight batches of the herbal samples were analyzed,the characteristic peaks were extracted,and hierarchical clustering analysis(HCA) and principle component analysis(PCA) were applied to classify and differentiate the samples.Result;The contents ofβ,β’dimethylacrylalkannin in all the samples were higher than the required limit in Chinese Pharmacopoeia.As the results of HCA and PCA analyses,all the samples could be clearly separated and classified into three groups,namely Arnebiae euchroma Radix,the imported Aniebiae Radix with unknown species and Onosmae Radix.Some crucial markers besidesβ,β’-dimethylacrylalkannin were found to be important in their quality evaluation.Conclusion;The commercial samples of Arnebiae Radix arenot the same in both the species and the qualities.The established HPLC method combined with multivariate statistical analysis can differentiate the samples,and can be further developed for the comprehensive quality evaluation of Arnebiae Radix.
