摘要
目的:建立测定人血浆中阿塞那平含量的HPLC法。方法:色谱柱:Diamonsil C_(18)(250 mm×4.6 mm,5μm)反相柱;柱温:25℃;流动相:乙腈-甲醇-缓冲液(15:55:30);流速:1.0 ml·min^(-1);检测波长:254 nm;内标:氯氮平。结果:阿塞那平血浆浓度在50~5 000μg·L^(-1)范围内线性关系良好(r=0.999 1),平均提取回收率为87.32%,日内RSD小于4.86%;日间RSD小于8.00%。结论:该方法快速、简便,结果准确,可以用于阿塞那平的临床研究。
Objective:To establish a RP-HPLC method for the determination of asenapine in human plasma.Method;A Diamonsil? C18(250 mm×4.6 mm,5μm) column was adopted.The column temperature was set at 25℃.The mobile phase was acetonitrilemethanol -buffer(15:55:30) with a flow rate of 1.0 ml·min-1.The UV detection wavelength was set at 254 nm.Clozapine was used as internal standard to determine asenapine.Result:The linear calibration curve was obtained within the concentration range of 50~5000μg·L-1(r = 0.999 1).The average extraction recoveiy was 87.32%.The intra-day RSD was less than 4.86%and inter-day RSD was less than 8.00%.Conclusion:The method is rapid,convenient and accurate,and can be used in the clinical research on asenapine.
