内部沸腾法提取银叶树叶总黄酮的研究 被引量:16

摘要 为优化银叶树叶中总黄酮的内部沸腾法提取工艺,以提高提取物的黄酮产率及含量,采用单因素试验的方法,考察解吸及提取过程中的解吸剂浓度、解吸剂用量和解吸时间及提取剂浓度、提取温度、提取时间和提取剂用量等7个因素对提取效果的影响,结果表明,银叶树叶先用浓度80%乙醇3 mL/g解吸20 min,然后用浓度30%乙醇25 mL/g在85℃的条件下提取两次,每次5 min,在该工艺条件下总黄酮提取得率为13.5%,提取物的黄酮含量达到46.9%。与传统的醇提法及酶提法相比,内部沸腾法提取时间分别缩短了70 min和140 min,乙醇用量分别减少了46 mL和15 mL,但总黄酮提取率比前两种方法分别提高了4.8%和2.8%,黄酮含量分别提高了14.5%和3.1%。内部沸腾提取法快速、有效且经济,用于银叶树叶中总黄酮的提取效果良好。 In order to improve the yield and content of product and optimize the extraction technology of the total flavonoids in leaves of Heritiera littoralis by inner ebullition, single factor experiments were conducted to investigate the effects of seven factors on the extraction, including the concentration and the amount of desorption agent, desorption time, the concentration and the amount of extractant, extraction temperature and time. It was found that when leaves of Heritiera littoralis were desorbed 20 minutes using 80% ethanol 3 mL/g and then extracted twice (5 min/time) in 30% ethanol 25 mL/g under 85℃, the yield of total flavonoids of 13.5% was obtained and the content of tlavonoids in extractive reached 46.9%. Compared with the traditional ethanol and cellulase extraction methods, the inner ebullition extraction had the following obvious advantages that extraction time was reduced by 70 min and 140 min, ethanol consumption reduced by 46 mL and 15 mL, while the yields of total flavonoids increased by 4. 8% and 2. 8% , and the contents of flavonoids in extractive raised by 14.5% and 3.1% ,respectively. Inner ebullition extraction was rapid, effective and economic in the extraction of total flavonoids from leaves of Heritiera littoralis.
