摘要
为探索药用植物牛耳枫Daphniphyllum calycinum的综合利用价值,以褐飞虱Nilaparvata lugens、水稻纹枯病菌Rhizoctonia solani、番茄白绢病菌Sclerotium rolfsii、香蕉枯萎病菌Fusarium oxysporum f.sp.cubense以及粉纹夜蛾Trichoplusis ni卵细胞系(Hi-5细胞系)为靶标生物,测定了牛耳枫叶的甲醇粗提物及其萃取物的生物活性。结果表明,乙酸乙酯萃取物对上述3种植物病原菌和Hi-5细胞系均表现出显著的生物活性。采用柱层析分离技术和重结晶等方法从活性部位乙酸乙酯萃取物中分离得到3个化合物,经核磁共振氢谱、碳谱解析分别鉴定为反式对甲氧基肉桂酸(化合物1)、木犀草素(化合物2)和槲皮素(化合物3)。其中化合物1和化合物2为首次从该属植物中分离得到。化合物2对Hi-5细胞系具有显著的细胞毒活性,48 h的EC50值为11.26μg/mL。研究结果为牛耳枫在植物源农药领域的开发利用提供了理论依据。
In order to explore the comprehensive utilization of Daphniphyllum calycinum Berth, bioactivities of the methanol extract and its solvent extracts from the leaf of D. calycinum against Ni- laparvata lugens, Rhizoctonia solani, Sclerotium rolfsii, Fusarium oxysporum f. sp. cubense, as well is ni cells ( Hi-5 cells). The results showed that the bioaetivities against three plant pathogens and Hi-5 cells. Three active EtOAe-soluble fraction by column chromatography and rec EtOAc-soluble fraction had obvious compounds were isolated from the rystallization. Their structures were identified as trans-p-methoxy-cinnamic acid ( compound 1 ), luteolin ( compound 2 ), quercetin ( compound 3) by 1H NMR and 13 C NMR analysis. Compounds 1 and 2 were obtained from the genus Daphniphyllum for the first time. Compound 2 exhibited potent eytotoxicity against Hi-5 ceils, with ECs0 value of 11.26μg/mL after treatment in 48 h. The study provided theoretical basis for utilization of D. calycinum in the area of botanical pesticides.
