摘要
目的:检测K14-VEGF转基因小鼠皮肤基因组内银屑病相关细胞因子表达水平。方法:通过实时(real-time)PCR和免疫组化的方法,测定VEGF、IL-6、IL-17、Stat-3等细胞因子mRNA与Stat-3蛋白的表达。结果:转基因小鼠皮肤中银屑病相关细胞因子Stat-3mRNA、VEGF mRNA以及Stat-3蛋白表达水平与野生型小鼠相比显著增高(P<0.05,P<0.05,P<0.01)。结论:K14-VEGF转基因小鼠皮肤基因组内上述银屑病相关细胞因子表达上调。
Objective:To detect the expression levels of cytokines related to psoriasis in K14-VEGF transgenic mouse skin.Methods:Gene expression of cytokines(VEGF,IL-6,IL-17,Stat-3)and Stat-3protein expression were detected by real-time PCR and immunohistochemistry,respectively.Results:Stat-3mRNA,VEGF mRNA and Stat-3protein expression levels in the skin of transgenic mouse were significantly higher,compared with those of wild mice(P0.05,P0.05,P0.01).Conclusion:The expression levels of cytokines related to psoriasis(Stat-3,VEGF)were up-regulated in K14-VEGF transgenic mouse skin.
