摘要
目的观察不同时程低氧诱导的肺动脉高压小鼠肺组织中IL-17mRNA及其蛋白表达水平的变化,探讨IL-17在低氧诱导肺动脉高压中的作用。方法建立小鼠低氧诱导肺动脉高压模型,监测小鼠血流动力学改变、右心室肥厚指数(RV/LV+S),RT-PCR技术检测对照组及低氧3周、6周组肺组织IL-17mRNA表达水平变化,Western blot技术检测相应蛋白表达水平变化及免疫组化方法对IL-17进行定位。结果与对照组相比,低氧诱导下小鼠右心室收缩压力及右心室肥厚指数均明显升高,且差异有统计学意义(均P<0.01),证实肺动脉高压模型成功建立。随着低氧时间的延长,IL-17mRNA及其蛋白表达水平也逐渐增加,差异均有统计学意义(P<0.05或P<0.01)。IL-17主要定位于内皮细胞,肺泡上皮细胞也有表达。结论 IL-17可能在低氧诱导肺动脉高压发生、发展过程中扮演了重要角色。
Objective To observe the changes of IL-7 mRNA and protein expressions in mouse pulmonary hypertension models induced by hypoxia for different time in an attempt to examine the role of IL-17 in hypoxia-induced pulmonary hypertension.Methods The mice were exposed to hypoxia for 3 or 6 weeks to establish hypoxia-induced pulmonary hypertension models.The hemodynamic changes of the mice were monitored.Right ventricular systolic pressure(RVSP)was measured by a right cardiac catheterization and right ventricular hypertrophy index[right ventricle to left ventricle plus interventricular septum(RV/LV+S)]was obtained.The pulmonary IL-17 mRNA expression was detected by RT-PCR and the IL-17 protein expression by Western blot.Immunohistochemical staining was used to examine the location of IL-17 in mouse pulmonary tissues.Results The RVSP and RV/(LV+S)were much higher in hypoxia-induced groups than in control group(P〈0.01),suggesting the success of the model establishment.The mRNA and protein expression levels of IL-17 were significantly increased with hypoxia time(P〈0.05 or P〈0.01).IL-17 mainly located in the endothelial cells,also in the alveolar epithelial cells.Conclusion IL-17 may play an important role in hypoxia-induced pulmonary hypertension.
