朽木中白腐真菌的选育及对木质纤维素降解性能研究 被引量:1

摘要 从自然界腐朽木材上分离筛选出白腐真菌,并对其降解木质纤维素的能力进行了分析.通过选择培养基富集培养、PDA培养基划线分离初步筛选菌株,利用显微镜观察、生长曲线的变化及木质素氧化酶的显色反应复筛,通过滤纸条崩解实验以及稻草固体发酵实验分析白腐真菌的木质纤维素降解能力.结果表明,选育出的B2菌与E5菌株形态特征符合白腐真菌的生物特性,且存在木质素氧化酶,培养第6天菌株对应的滤纸失重率可达31.04%和25.59%,固体发酵20天后木质素降解效率为33.7%和30.7%.B2菌和E5菌株均具有较强的降解木质纤维素的能力,且B2菌株的降解效果优于E5菌株. The research purposes is to isolate and screen white rot fungi from the nature decayed wood, and analyse the ability to degrada lignocellulose. Using the selective medium en- riched culture,PDA medium crossed separation preliminarily screened strains. Using microscope, the change of growth curve and the chromogenic reaction of lignin oxidase compound rescreened strains. The lignocellulose degradation ability of white rot fungi was analysed by the filter paper disintegration test and straw solid state fermentation test. The results show that the morphological characteristics of bred B2 and E5 strains is in line with the biological characteristics of white rot fungi. Both of them have lignin oxidase and their filter weight loss rate can respectively reach 31. 040/oo and 25.59; after cultivated 6 days,lignin degradation ef- ficiency up to 33. 7% and 30. 7% after solid-state fermentation 20 days. B2 and E5 strains have the strong ability to degrade lignocellulose. The degradation effect of B2 strain is better than that of E5 strain.
