摘要
目的探讨PD-L2在人胎盘间充质干细胞黏附和迁移过程中的生物学意义。方法应用消化法分离、培养人胎盘间充质干细胞,体外扩增培养3代后用于实验;RT-PCR检测PD-L2在人胎盘间充质干细胞上的表达;应用化学合成的PD-L2 siRNA阻断PD-L2在人胎盘间充质干细胞上的表达;细胞计数法检测人胎盘间充质干细胞的黏附能力;Transwell法检测人胎盘间充质干细胞的迁移能力。结果人胎盘间充质干细胞高表达PD-L2分子,PD-L2 siRNA能有效阻断人胎盘间充质干细胞对PD-L2的表达;细胞计数结果显示,接种后0.5 h阻断组人胎盘间充质干细胞的黏附率与正常对照组相比无显著升高,在接种后1 h和3 h,阻断组人胎盘间充质干细胞的黏附率均明显高于正常对照组;Transwell检测结果显示,与正常对照组相比,在DMEM-LG、含SDF-1α的DMEM-LG、人胎盘间充质干细胞培养上清三种培养条件下,阻断组细胞迁移率均显著降低。结论 PD-L2表达在人胎盘间充质干细胞,且在人胎盘间充质干细胞的黏附和迁移中发挥重要作用。
Objective Discussed the PD - between L2 in human placenta mesenchymal stem cell adhesion and mi- gration in the process of biological significance. Methods The digestion method between placenta mesenchymal stem cells were isolated and cultured, in vitro amplification culture after three generations used in experiments. Results The placental mesenchymal stem cells between high expression PD - L2 molecules, PD - L2 siRNA can effectively block between placenta mesenchymal stem cells in PD - the expres- sion of L2. Cell count, according to the results of vaccination after 0.5 h block group between placenta adhesion rate of mes- enchymal stem cells compared with normal control group no significant increases, in the 1 h and 3 h after inoculation, block- ing group between placenta adhesion rate of mesenchymal stem cells were significantly higher than normal control group; Transwell test results showed that compared with normal control group, in DMEM - LG, DMEM containing alpha SDF - 1 - LG, people between the placenta mesenchymal stem cell culture supematant of three conditions, cell mobility of block- ing group were significantly reduced. Conclusion The PD- between L2 expression in human placenta mesenchymal stem cells, and between the people of the placenta adhesion and migration of mesenchymal stem cells play an important role.
