金水鲜胶囊联合放射治疗对肺腺癌A549细胞抑制率和VEGF表达影响的研究 被引量:5

Influence of Goldwater Fresh Capsule Combined with Radiotherapy on Inhibition Rate and VEGF Expression of Lung Adenocarcinoma A549 Cells

摘要 [目的]研究金水鲜胶囊联合放疗对肺腺癌A549细胞增殖的抑制作用,同时检测采用两者联合治疗对肺癌细胞分泌的VEGF蛋白及mRNA表达的影响。[方法]体外培养肺腺癌A549细胞,将处于对数期的A549细胞随机分为4组:空白对照组(NC)、金水鲜组(JSX)、放疗组(RT)以及金水鲜与放疗联合组(JSX+RT),其中联合组又按照时序不同分为3个亚组:先金水鲜后放疗组(JSX→RT),先放疗后金水鲜组(RT→JSX),同时金水鲜联合放疗组(JSX+RT)。MTT法计算细胞生长抑制率,ELISA法测定VEGF蛋白表达的含量,RT-PCR技术检测各处理组VEGFmRNA的表达变化。[结果]①与对照组相比,各处理组肺腺癌细胞的生长速度随时间的改变都受到不同程度的抑制,其中以JSX+RT组最为明显,RT组其次,JSX组抑制肺腺癌细胞的增殖程度不如其他两个实验组。JSX+RT组抑制细胞增殖程度与空白对照组、JSX组、RT组相比,均具有显著统计学差异(P<0.05)。②各处理组细胞表达的VEGF蛋白含量有所下降,其中JSX组及JSX+RT组在48h时VEGF蛋白表达量下降的作用明显,与空白对照组及RT组相比,差异有显著统计学意义(P<0.05)。③与空白对照组相比,各处理组VEGFmRNA的表达均表现出下降趋势,在48h时JSX组及JSX+RT组与空白对照组、RT组相比,差异有显著统计学意义(P<0.05),RT组与空白对照组相比,差异有明显统计学意义(P<0.05)。[结论]金水鲜联合放疗可显著抑制肺腺癌A549细胞增殖,下调VEGF蛋白以及mRNA的表达,间接降低新生血管的生成速度和数量,考虑为金水鲜胶囊在影响血管生成方面能增加放疗对肺癌的杀伤作用。
