星点设计-响应面法优选芪玄抑甲宁的提取工艺 被引量:8

摘要 目的:优选芪玄抑甲宁方中有效成分的提取工艺。方法:以浸泡时间、加水量、提取时间和提取次数为自变量,毛蕊异黄酮葡萄糖苷、哈巴苷和迷迭香酸得率为因变量,通过对自变量各水平的多元线性回归及二项式拟合,采用星点设计-响应面法选取最佳工艺,并进行预测分析。结果:最佳提取工艺条件为浸泡时间74.4min,加水量为13.56倍,提取时间1.165h,提取次数2次,在此条件下,芪玄抑甲宁中毛蕊异黄酮葡萄糖苷、哈巴苷、迷迭香酸含量总和的最大估计值为0.36,与预测值相符。结论:该方法简便合理,稳定,可预测性较好。 Objective: To optimize extraction technology of Qixuan Yijianing.Methods: With soaking time,the amount of water,extraction time and extraction times as independent variables,yields of calycosin-7-O-β-D-glycoside,harpagide and rosmarinic acid as dependent variable,multiple linear regression and binomial of each level of independent variableswere fitted.Optimum technology was selected by central composite design and response surface methodology,and prediction was carried out by comparing the observed and predicted values.Results: Optimum extraction technology parameters were as follows: soaking time 74.4min,the amount of water 13.56-fold,extraction time 1.165h and extraction times twice.Under this condition,maximum estimated value of the sum of calycosin-7-O-β-D-glycoside,harpagide and rosmarinic acid was 0.36,which was agreed with model predicted value.Conclusion: Optimized extraction technology is simple,reliable and highly predictive.
