Study on Three Sets of Cytoplasmic Male Sterility Cotton from Different Countries:Locate the Molecular Markers of Restoring Genes

摘要 【目的】研究定位出来源于不同国家的棉花细胞质雄性不育恢复基因的分子标记,为棉花三系杂交育种提供恢复系分子标记辅助选育技术。【方法】对国内、以色列和美国引进的三套棉花胞质雄性不育系及相应恢复系,通过构建的三套BC_1育性BSA-DNA池,利用1 000对SSR和STS、RAPD等分子标记的分析。【结果】来源于美国引进恢复系含恢复基因有2个,而来源于国内与以色列引进的恢复系,只含有1个恢复基因。定位出与恢复基因紧密连锁的两侧标记(美国、国内、以色列)分别为:Rf_1位于BNL3535、CIR179间,两标记间5.3 cM。Rf_2位于STS659、BNL1045间,两标记间4.8 cM;Rf位于CIR222、BNL632间,两标记间6.7 cM;Rf位于STS147、CIR179间,两标记间4.3 cM。【结论】初步获得三套来源于不同国家的细胞质雄性不育恢复基因的分子标记图谱。 Abstract:[ Objective ] The main purpose of this research is to study sets of cytoplasmic male sterility cotton from different countries and locate the molecular markers of restoring genes in the hope of providing seed selection techniques for restoring line - assisted selection by the technology of molecular maker in cotton three lines hybrid breeding. [ Method] Three sets of BSA -DNA pools of BC1 fertility were constructed by three sets of cotton cytoplasmic male sterility lines corresponding to restorers from China, Israel and the United States, whieh were analyzed by 1 000 suits molecular markers of SSR, STS, and RAPD. [ Result] Three sets of molecular marker maps linkage closely with restoring gene were obtained : there are two restoring genes from restorers that were derived from the United States, but only one restoring gene in restorers was derived from China and Israel. Marks (from the United States:R fl and Rf2, China and Israel: Rf ) on both sides of the linkage closely restoring genes were separately located: Rfl lies in between BNL3535 and CIR179, in which the distance was 5.3 cM;Rf2 lies in between STS659 and BNL1045, in which the distance was 4.8 cM;Rflies in CIR222 and BNL632,in which the distance was 6.7cM;and Rf lies in distance was 4.3 cM. [ Conclusion ] The molecular maker gene map of the three sets of cytoplasmic male sterility cotton from different countries has initially taken shape
