摘要
目的探讨卵巢组织中结蹄组织生长因子(CTGF)、肝细胞生长因子(HGF)及间隙连接蛋白(Cx-43)水平在多囊卵巢综合征(PCOS)患者内的变化,同时对患者进行干预,寻求较为有效的治疗手段。方法选择2012年2~12月就诊于湖北医药学院附属太和医院的PCOS患者60例,将其作为PCOS组,另选择同期健康体检者60例作为对照组,对PCOS组患者行药物治疗干预,每天口服达英-35,连续服用21 d。利用显微镜对研究对象进行卵巢形态学的观察,CTGF、HGF、Cx-43水平测定采用双抗体夹心法。结果卵巢形态学观察结果显示,对照组多个不同发育程度的卵泡形态完整,PCOS组卵泡呈扩张状,且排列松散,经干预,PCOS组颗粒细胞层数有所增加,形态趋于完整。与对照组比较,PCOS组干预前颗粒细胞及卵泡膜细胞中,CTGF[(9.01±2.31)μg/L]、HGF[(9.88±3.21)μg/L]水平显著升高(P<0.05),Cx-43[(11.1±4.3)μg/L]表达水平降低(P<0.05)。经干预,PCOS组CTGF[(6.32±1.17)μg/L]、HGF[(7.13±2.56)μg/L]水平降低(P<0.05),Cx-43[(13.4±5.4)μg/L]表达水平升高(P<0.05)。结论 CTGF、HGF、Cx-43表达在PCOS的高胰岛素血症、高雄激素血症的形成和无排卵中起着重要的作用,可选择达英-35对PCOS患者进行干预治疗。
Objective To study the change of CTGF, HGF, Cx-43 in ovarian tissue in patients with CTGF, to seek more effective treatments by giving intervention. Methods 60 patients with PCOS from February to December 2012 in clinic in Taihe Hospital Affiliated to Hubei Medical University were chosen as PCOS group, the other options of 60 cases in the same period with health check-up were chosen as control group, and were given the intervention of drug therapy in patients with PCOS group, Diane-35 was given by oral a day with 21 d. Microscope was used to ovarian morphology observation of the object of study, CTGF, HGF, Cx-43 level measurement were detected with double antibody sandwich method. Results Results of ovarian morphology observation showed, the control group of different follicular development degree of configuration was complete, the PCOS group follicle was expanding, and their arrangement was loose, after the intervention, granular cell layer increased, with form of complete. Compared with control group, the PCOS group before intervention in granulosa cell and theca cells, CTGF [(9.01±2.31) μg/L], HGF [(9.88±3.21) μg/L] were significantly increased (P 〈 0.05), Cx-43 [(11.1±4.3)μg/L] expression level decreased (P 〈 0.05). After the intervention, CTGF in PEOS group [(6.32±1.17) μg/L], HGF [(7.13±2.56) μg/L] levels were lower (P 〈 0.05),Cx-43 [(13.4±5.4) μg/L] expression level increased (P 〈 0.05). Conclusion CTGF, HGF, Cx-43 expression of high androgen levels in PCOS hyperinsulinemia and anovulatory plays an important role, and Diane-35 can be chosen for intervention therapy in patients with PCOS.
