九味白术汤对实验性溃疡性结肠炎大鼠血清白介素-1β及白介素-10的影响 被引量:8

摘要 目的观察九味白术汤(JWBZ)对溃疡性结肠炎(UC)大鼠血清白介素-1β(IL-1β)及白介素-10(IL-10)水平的影响。方法 48只大鼠完全随机分为正常对照组、模型组、柳氮磺胺吡啶(SASP)组、JWBZ高剂量组、中剂量组与低剂量组;采用2,4,6-三硝基苯磺酸(TNBS)复制大鼠溃疡性结肠炎模型;连续给药10天后,观察UC大鼠结肠黏膜损伤指数(CMDI)、血清IL-1β及IL-10水平的变化。多组间比较采用单因素方差分析。结果正常对照组、模型组、JWBZ低、中、高剂量组及SASP组的CMDI分别为:(0.25±0.46)、(3.38±1.06)、(1.63±0.92)、(1.50±0.53)、(2.50±0.93)、(2.38±0.74)分;相应的IL-1β水平分别为:(232.94±21.77)、(756.67±71.70)、(469.57±71.13)、(401.97±39.24)、(598.93±45.34)、(655.66±10.51)ng/L;IL-10水平分别为:(52.14±7.07)、(20.19±5.17)、(56.79±9.47)、(63.96±9.37)、(41.73±10.55)、(51.13±2.81)ng/L。与正常对照组比较,模型组CMDI显著增加(P<0.01),血清IL-1β水平显著升高(P<0.01),而血清IL-10水平显著降低(P<0.01);与模型组比较,JWBZ低、中、高剂量组及SASP组CMDI均显著降低(P<0.05或P<0.01),血清IL-1β水平均显著降低(P<0.05或P<0.01),而血清IL-10水平显著升高(P<0.01);与SASP组比较,JWBZ中剂量组CMDI显著降低(P<0.05),高、中剂量组血清IL-1β水平显著降低(P<0.01);中剂量组血清IL-10水平显著升高(P<0.01),而低剂量组血清IL-10水平显著降低(P<0.05)。结论 JWBZ对UC大鼠结肠损伤具有良好的修复作用,其机制可能通过调节促炎因子与抗炎因子的平衡实现。 Objective To observe the effect of Jiuweibaizhutang (JWBZ) on serum levels of interleukin-1β(IL-1β) and interleukin-10 (IL-10) in ulcerative colitis (UC) rats. Methods 48 rats were randomly divided into 6 groups: norma! group, model group, salazosulfapyridine (SASP) group, low-dose, middle-dose and high-dose JWBZ groups. Ulcerative colitis rat model was copied by 2, 4, 6-TNBS. After 10 days'treatment, the changes of colonic mucosal damage index (CMDI), serum levels of IL-1β and IL-10 were observed. Significant differences between groups were tested by one-way analysis of variance (ANOVA). Results The CMDI of normal group, model group, low-dose, middle-dose and high-dose JWBZ groups were(0.25±0.46)、(3.38±1.06)、(1.63±0.92)、(1.50±0.53)、(2.50±0.93)、(2.38±0.74)scores,respectively; levels of IL-1β were(232.94±21.77)、(756.67±71.70)、(469.57±71.13)、(401.97±39.24)、(598.93±45.34)、(655.66±10.51)ng/L,respectively; and level of IL-10 were(52.14±7.07)、(20.19±5.17)、(56.79±9.47)、(63.96±9.37)、(41.73±10.55)、(51.13±2.81)ng/L,respectively. Compared to normal group, the CMDI of model group was significantly increased (P 〈 0.01), and the serum IL-1β level of it increased significantly (P 〈 0.01), while IL-10 level decreased significantly (P 〈 0.01). Compared to model group, the CMDI of JWBZ and SASP groups decreased significantly (P 〈0.05 or P 〈 O.01), and the serum IL-II3 level of treatment groups decreased significantly (P 〈 0.05 or P 〈 0.01), while the IL-10 level increased significantly (P 〈 0.01). Compared to SASP group, the CMDI of middle-dose JWBZ group significantly decreased (P 〈 0.05), the serum IL-1β level of high-dose and low-dose JWBZ groups decreased signifi- cantly (P 〈 0.01), the serum IL-10 level of middle-dose JWBZ group increased significantly (P 〈 0.01), while it decreased significantly of low-dose JWBZ group (P 〈 0.05). Conclusion JWBZ treated UC rats effectively by regulating the balance of pro-and anti-inflammatory cytokines.
