摘要
目的探讨含利塞膦酸磷硅酸钙水门汀浸提液对胎鼠成骨细胞增殖、分化和功能的影响。方法利用MTT法检测含利塞膦酸磷硅酸钙水门汀浸提液对成骨细胞增殖影响,用酶标仪检测碱性磷酸酶(ALP)比活性。结果添加0.1%和0.5%利塞膦酸的骨水泥对成骨细胞没有毒性,1%利塞膦酸的骨水泥浸提液与成骨细胞共同培养3d后对成骨细胞产生毒性反应。0.1%和0.5%利塞膦酸浸提液对成骨细胞ALP比活性增加。结论高剂量含利塞膦酸的磷硅酸钙水门汀对成骨细胞毒性较小,是潜在的填充骨缺损的新型材料。
Objective To explore the effect of phosphorus-containing silicate cements risedronate acid extracts on the osteoblast proliferation,differentiation and function of fetal rat. Methods Using the MTT assay to detect the effect of containing phosphorus silicate cements risedronic acid extracts on osteoblast proliferation,and to assay the alkaline phosphatase activity using a microplate reader detection test. Results Addition of 0. 1% and 0. 5% risedronate acid bone cement was not toxic on osteoblast,whereas 1% of the bone cement risedronie acid extracts together with osteoblasts cultured for 3 days produce toxicity on osteohlast. ALP activity was increase in 0.1 % and 0. 5% of risedronate acid extracts group. Conclusion Risedronate dose containing calcium silicate cements phosphorous acid osteoblast was relative low toxicity, which was a potential new bone defect filling material in the future.
