摘要
目的初步验证掺锶硫酸钙复合骨修复材料修复松质骨骨缺损的能力。方法建立直径5mm、深度12mm的新西兰兔股骨髁包容性骨缺损模型,取含锶量为0.5%的掺锶硫酸钙骨缺损进行修复。术后4、8、12w连续X线摄片,观察骨缺损修复情况。术后12w处死动物,取动物双股骨下段,进行股骨远端缺损区压凹实验。术后8、12w取标本行HE染色,光镜观察材料及周围组织的变化和植入材料内新骨的形成情况。采用ImageplusCCD系统对植骨区的成骨情况进行定量分析。结果 X线显示掺锶硫酸钙组术后12w时原骨缺损区域已完全被新骨填充,其密度与正常骨组织密度接近;α-半水硫酸钙组术后12w时骨缺损区内部仍可见少量植入材料的碎片。术后12w掺锶硫酸钙组和α-半水硫酸钙组股骨髁骨缺损区压缩强度均高于空白对照组(P=0.000,P=0.000);两组间比较掺锶硫酸钙组压缩强度高于α-半水硫酸钙组(P=0.007);掺锶硫酸钙组压缩强度与正常对照组相比差异无统计学意义(P=1.000)。术后8、12w掺锶硫酸钙组和α-半水硫酸钙组新骨形成率均明显高于空白对照组(P=0.000),且掺锶硫酸钙组新骨形成率均明显高于α-半水硫酸钙组(P=0.030,P=0.043)。结论现阶段制备的0.5%掺锶硫酸钙材料在松质骨包容性骨缺损区内有明显的促成骨作用,在12w内比基质材料α-半水硫酸钙能更好的修复骨缺损。
Objective To determine whether the strontium calcium sulfate compound can be used as a new bioactive material for cancellous bone defect repairing in clinic or not. Methods The compound contains 0.5% SrC12 was used to repair bone cavity defect model,a condyles of femur of 5 mm in diameter and 12 mm in depth. 4,8 and 12 weeks after the surgery, the repair of defected bones were assessed by using X-ray. On the 12th week post surgery, mechanical test was performed. The newly formed tissues in defected areas were assayed by H. E. staining. Results The results of X-ray showed that 12 weeks after operation, the defected bone was completely filled with new bone tissue in the strontium-calcium sulfate group, and the density of new tissues was similar to the normal bone. However,there had some fragments of the implantation in the a CSH group at the 12th week post operation. Anyhow,situation in the both groups were significantly better than that in the control group ( P = 0. 000, P=0. 000). The compressive strength in strontium-calcium sulfate group was nearly the same with that in the control group (P=1.000),however,significantly higher than that in the a-CSH group (P=0. 007). Bone formation rate was also higher that in a-CSH group (P = 0. 030, P 0. 043) and control group (P = 0. 000). Conclusion The strontium calcium sulfate compounds containing 0.5 ~//00 SrCI2 can repair large cancellous bone cavity defect better.
