神经生长因子对猪卵丘细胞扩散及卵母细胞体外成熟的影响

Effect of Nerve Growth Factor(NGF) on Pig Cumulus Cell Expansion and Oocyte in vitro Maturation

利用光学显微镜观察及地衣红染色方法,探究猪卵母细胞体外成熟过程中添加神经生长因子(NGF)对猪卵丘细胞扩散和卵母细胞成熟的影响。结果表明:在猪卵母细胞体外成熟过程中添加NGF对猪卵丘细胞扩散无明显影响;单纯添加NGF并不能显著改变第二次减数分裂中期(MⅡ期)卵母细胞的比率,但却可以显著提高生发泡期(GV期)卵母细胞比率。当NGF与促性腺激素(eCG和hCG)联合作用时,MⅡ期卵母细胞比率相对于单纯地促性腺激素作用有显著提高。

The aim of this work is to assess the effect of nerve growth factor on pig cumulus expansion and in vitro maturation（IVM） of pig oocytes.The degree of cumulus expansion was assessed directly under light microscope while nuclear maturation was assessed in terms of orcein staining.The results showed that NGF could not stimulate expansion of cumulus cells.Compared with oocytes in control group,oocytes stimulated with NGF alone were more likely to stay at GV phase,although the percentage of oocytes that had reached metaphase Ⅱ phase was not significantly different.Nevertheless,oocytes stimulated with NGF combined with gonadotrophins（equine chorionic gonadotropin and human chorionic gonadotropin） were more likely to reach metaphase Ⅱ phase compared with those stimulated with gonadotrophins alone.
