安脑平冲片对大鼠脑出血后血肿周围组织AQP-9表达的影响 被引量:2

摘要 目的通过观察大鼠脑出血后血肿周围组织内水通道-9(aquaporin-9,AQP-9)的表达变化的影响来探讨安脑平冲片对脑出血的保护机制。方法采用自体动脉血注入法制作大鼠脑出血模型。大鼠随机分为正常组、假手术组、模型组、安脑平冲片组、甘露醇组、联合用药组,各组分别给药,于脑出血后24、48、72 h处死开颅取脑,采用干湿称质量法测定脑组织含水量、免疫组化法测定AQP-9的表达。结果 (1)大鼠脑出血后血肿周围脑组织含水量逐渐增加(P<0.05)。安脑平冲片组、甘露醇组、联合用药组各时间点与模型组比较脑组织含水量均相应降低,差异具有统计学意义(P<0.05)。其中给药后联合用药组72 h效果最佳。(2)大鼠脑出血后模型组各时间点AQP-9表达均显著增加(P<0.01)。与模型组比较,安脑平冲片组、联合用药组各时间点AQP-9表达均降低(P<0.01);与甘露醇组比较无统计学意义。联合用药组与安脑平冲片组比较于48、72 h时AQP-9表达降低更显著(P<0.05)。结论安脑平冲片可能通过减少脑组织内AQP-9的表达水平,以减轻脑水肿保护脑组织。 Objective To investigate the protection mechanism of Annao Pingchong pills on intracerebral hemorrhage through the observation of the expression changes of peripheral tissue aquaporin 9 after intracerebral hemorrhage in rats.Methods The intracerebral hemorrhage model in rats were established by injecting autologous arterial blood into caudate nucleus.The rats were randomly divided into sham-operation group,model group,mannitol group,Annao Pingchong pill group and combined treatment group.All rats were administrated and the brain specimens were collected.Water contents in brain tissues were tested using dry and wet weight method and the expression levels of AQP-9 were detected by the immunohistochemistry after intracerebral hemorrhage 24 h,48 h and 72 h.Results ①Compared with sham-operation group,the water contents in brain tissue around the hematoma gradually increased in rats with intracerebral hemorrhage(P0.05).Compared with the model group in each time point,the water contents of Annao Pingchong pill group,mannitol group and combined treatment group were reduced accordingly,and the differences were statistically significant(P0.05).Annao Pingchong pill combined mannitol group was best at 72 hours.②The expression levels of AQP-9 were increased in each time point in rats with intracerebral hemorrhage(P0.01).Compared with the model group,the AQP-9 expression levels of Annao Pingchong pill group and combined treatment group were reduced remarkably(P0.05) in each time point,but there was no significant difference in mannitol group.Compared with Annao Pingchong pill group,the AQP-9 expression levels of combined treatment group were markedly decreased at 48 h and 72 h.Conclusion Annao Pingchong pills protect the brain tissues by alleviating brain edema and decreasing AQP-9 expression.
