川芎嗪对抗油酸致大鼠急性肺损伤的机制

摘要 目的研究川芎嗪(TMP)对油酸致大鼠急性肺损伤(ALI)的保护作用及其可能机制。方法采用注射油酸(OA)复制ALI大鼠模型。将36只雄性SD大鼠随机分为3组:对照组、油酸组和川芎嗪组。各组大鼠均于注射油酸或生理盐水后1 h和4 h处死并观察。光镜下观察肺、肝组织形态学改变;检测各组大鼠丙二醛含量和超氧化物歧化酶活性的表达变化。结果肺、肝组织形态学变化提示川芎嗪组大鼠肺组织炎症反应较油酸组减轻。随着时间延长,油酸组和川芎嗪组大鼠MDA含量增加(P<0.05)、SOD活性降低(P<0.05),而对照组二者均无明显变化(P>0.05)。相同时间点川芎嗪组较油酸组MDA含量降低(P<0.05),但仍高于对照组(P<0.05);相同时间点川芎嗪组较油酸组SOD活性升高(P<0.05),但仍低于对照组(P<0.05)。结论川芎嗪可对抗油酸致大鼠ALI,其机制可能与减少ALI大鼠体内自由基水平有关。 Objective To investigate the protective effect of tetramethylpyrazine (TMP) to acute lung injury in rats induced by oleic acid and its possible mechanisms. Methods Rat models of acute lung injury induced by oleic acid (OA) were used. Thirty six Sprague-Dawley male rats were randomly divided into 3 groups: Control group, OA group and TMP group. All rats in each group were killed at lh and 4h after oleic acid infusion or physiological saline infusion. Morphological changes of lung tissues and liver tissues were observed under light microscope. The content of Malonaldehyde (MDA) and the activity of superoxide dismutase (SOD) were detected in each group. Results Morphological changes of lung tissues and liver tissues showed that pulmonary inflammation was relieved in TMP group compared with that of OA group. The content of MDA increased (P〈0.05) and the activity of SOD decreased (P〈0.05) progressively in OA group and in TMP group with the time prolonging, but the change was not obvious in control group (P〉0.05). The content of MDA in TMP group were lower than those in OA group at the same time (P〈0.05), but higher than that of the control group (P〈0.05). The activity of SOD in TMP group were higher than those in OA group at the same time (P〈0.05), but lower than that of the control group (P〈0.05). Conclusion Tetramethylpyrazine could antagonize acute lung injury induced by OA, which is probably through decreasing the level of free radical in rats.
