野胡萝卜籽挥发油碳氢部位对SD大鼠体内和体外的潜在致畸胎作用

摘要 用体外原代胚胎细胞培养和体内传统致畸胎试验研究了野胡萝卜籽挥发油碳氢部位的潜在致畸胎作用。体内试验检查了77只孕鼠和703只胎鼠。挥发油体外试验浓度为50、100和150μg/ml,体内试验为625.0,312.5和156.3mg/kg,分别用生理盐水和维生素A作为阴性和阳性对照。结果表明,在体外挥发油对大鼠胚胎中脑和肢芽细胞的分化无抑制作用:体内亦无母体毒性、胚胎毒性,对胎鼠亦无致畸胎作用,也不影响其生长发育。两种试验均提示挥发油对SD大鼠无致畸胎作用,且两种方法的结果是一致的。 The teratogenic potential effects of the volatile oil of Daucus carat L. seed were studied in Sprague-Dawley rats with in vitro embryo midbrain and limb bud cells culture and in vivo conventional teratological experiment. All of 77 pregnant rats and 703 fetuses were examinated in vivo. The compound was administered into the embryo cells and pregnant rats at the doses of 50, 100 and 150μg/ml in vitro and of 625.0, 312.5 and 156.3mg/kg in vivo, respectively. Fish liver oil and normal saline were separately used for the positive and negative control groups. The results showed that the compound did not inhibit the differentiation of embryo midbrain and limb bud cells, and had no toxic effects on maternity, embryo and fetal growth for any treatment group. Based on these data, both tests in vitro and in vivo suggest that the volatile oil does not show the teratological effects in Sprague-Dawley rats, and the result in vivo is closely consistent with that in vitro.
