摘要
本研究应用放射免疫测定法测定了HL-60细胞系和白血病患者单个核细胞(MNC)的铜锌超氧化物歧化酶(SOD-1)含量,应用流式细胞仪(FCM)同时检测了白血病细胞SOD-1含量与S期细胞比例的关系。结果显示:HL-60,AML,CML和B-ALL的MNC的SOD-1水平分别较正常组有明显升高(P<0.001);白血病患者经化疗获得完全缓解后,其WBC的SOD-1水平降低至正常以下,复发时SOD-1水平升高并明显超过正常值(P<0.001)。白血病细胞S期细胞的比例增加或减低,与SOD-1水平有一定关系。初步研究结果提示白血病细胞SOD-1水平增加,超氧阴离子自由基减少,表明其氧化-还原系统功能紊乱,该紊乱与白血病的发生发展可能有一定关系。白血病细胞SOD-1水平的检测对诊断分型和探索新的治疗途径有意义。
In this study levels of copper-zinc suproxide dismutase (SOD-1) in HL-60 cells and mononucleocytes (MNC) of patients with leukemia were determined by radioimmunoassay and were correlated to DNA content and percentage of S phase of leukemic cells. The results showed that the levels of SOD-1 in HL-60 cells and MNC of AML, CML and B-ALL were higher than those in normal MNC (P< 0.001). The levels of SOD-1 in WBC decreased when leukemia patients got completely remission and increased while leukemia relapsed. These results suggest that the increased SOD-1 levels and decreased anionic free radical of superoxygen demonstrate dysfunction of oxidio-reductive system which may have some relation to the leukemogenesis and development in leukemic cells. Determination of SOD-1 in leukemic cells may also have significant value in diagnostic typing and therapy.
