宁心通痹胶囊对大鼠心肌缺血及胸主动脉收缩的影响

Effects of Ningxin Tongbi Capsule on Myocardial Ischemia and Contraction of Thoracic Aorta in Rats

目的:研究宁心通痹胶囊对在体急性心肌缺血模型大鼠与离体主动脉收缩的影响。方法:结扎左冠状动脉以复制大鼠心肌缺血模型。实验分为假手术(等体积0.5%CMC-Na)、模型(等体积0.5%CMC-Na)、地奥心血康(60mg/kg)与宁心通痹高、中、低剂量(6.0、3.0、1.5g/kg)组。记录给药前后各时间点的心电图T波、ST段偏移量;硝基四氮唑蓝组化染色法测定心肌梗死区质量与梗死面积百分率。采用大鼠离体胸主动脉条,观察不同质量浓度宁心通痹胶囊(26.70×10-3、13.35×10-3、6.68×10-3、3.34×10-3、1.67×10-3g/ml)对大鼠胸主动脉收缩的影响。结果:高、低剂量宁心通痹胶囊能显著降低模型大鼠的心电图T波改变量(P<0.01或P<0.05);高、中剂量宁心通痹胶囊能显著降低模型大鼠的心电图ST段偏移量(P<0.05);高、中剂量宁心通痹胶囊能显著降低模型大鼠心肌梗死区质量与梗死面积百分率(P<0.01或P<0.05);在26.70×10-3、13.35×10-3、6.68×10-3g/ml质量浓度下,宁心通痹胶囊能明显舒张大鼠离体胸主动脉条(P<0.01或P<0.05)。结论:宁心通痹胶囊具有改善大鼠心肌缺血和舒张体外血管的作用。

OBJECTIVE： To observe the effects of Ningxin tongbi capsule on myocardial ischemia and contraction of thoracic aorta in rats. METHODS： Myocardial ischemia model was induced by the ligation of left coronary artery. Model rats were divided into 6 groups, i.e. sham operation group （constant volume of normal saline）, model group （constant volume of normal saline）, Di’ao xinxuekang capsule group （60 mg/kg） and Ningxin tongbi capsule high-dose, medium-dose and low-dose groups （6.0, 3.0, 1.5 g/kg）. S-T migration and T wave of electrocardiogram were detected before and after medication. The area of myocardial infarction was determined by NBT immunohistochemical staining method. The effects of different concentrations of Ningxin tongbi capsule （26.70×10-3,13.35×10-3,6.68×10-3,3.34×10-3,1.67×10-3 g/ml） on the contraction of thoracic aorta were observed by using isolated thoracic aorta strips of rats. RESULTS：High-dose and low-dose of Ningxin tongbi capsule could reduce the T wave of electrocardiogram（P〈0.01 or P〈0.05）. High-dose and medium-dose of Ningxin tongbi capsule could reduce the ST migration of electrocardiogram（P〈0.05）. High-dose and medium-dose of Ningxin tongbi capsule could relieved myocardial infarction weight and the percentage of infarct area（P〈0.01 or P〈0.05）. Ningxin tongbi capsule（26.7×10-3,13.35×10-3, 6.68×10-3 g/ml） had significant relaxing effect on the contraction of isolated thoracic aorta strips in rats.（P〈0.01 or P〈0.05） CONCLUSIONS：Ningxin tongbi capsule can improve myocardial ischemia and can relax the contraction of blood vessel in rats.
