无机三氧化聚合物对人牙乳头干细胞TNF-α和IL-1表达的影响

摘要 目的探讨无机三氧化聚合物(mineraltrioxideaggregate,MTA)对人牙乳头十细胞(stemcellsfroma·alpapillaeelkSCAPs)早期炎性蝌子TNF-a和IL-l表达的影响。方法酶消化法分离增养人SCAPs,经免疫磁珠筛选后,采川2mg/m1.MTA刺激1d、3d后,酶联免疫吸附法(enzymelinkedimmunosodentassay,ELlSA)检测其上清液中肿瘤坏死因子-a(tunlornecrosisfactor—a,TNF—01)、白细胞介素1(Interleukin—l,IL-1)的含量,采川实时定艟PCR(real—timeRT—PCR)检测各组细胞TNF-a、IL—la、1L-1B基因的表达水平。结果经MTA刺激ld和3d后,SCAh细胞上清液rfJTNF-a、浓度均较正常对照组低(P〈0.01),细胞对照组低(P〈0.01).结论MTA可抑制SCAPs早期炎性因子TNF-a 、IL—1的表达,通过降低细胞的炎性因子来释放来减轻组织的炎性反应。 Abstract Objective To determine the effects of mineral trioxide aggregate (MTA) on the expression of TNF-a and IL-1 in human stem cells from apical papilla (SCAPs). Methods SCAPs were isolated by magnetic activated cell sorting (MACS) and cultured in oL-MEM medium containing 2 mg/mL MTA for 1 day and 3 days. Enzyme linked immunosorbent assay (ELISA) was applied to examine the TNF-a and IL-1 in the culture supematant of SCAPs. Quantitative RT-PCR was used to determine the gene expression of TNF-a, IL-la, and IL-1β in SCAPs. Results The expression levels of TNF-a and IL-1 at both protein expression and mRNA transcription were inhibited (P 〈0.01) when the cells were treated with MTA. Conclusion MTA can inhibit the expression of early-stage inflammatory cytokines and relieve the inflammation via reducing the secretion of inflammatory cytokines.
