热休克蛋白70-2基因多态性与糖尿病肾病的相关性研究

摘要 目的探讨热休克蛋白70-2(+1538A/G)基因多态性与2型糖尿病肾病的关系。方法采用聚合酶链反应-限制性酶切片段长度多态性分析技术检测261例2型糖尿病患者(其中糖尿病肾病组130例,糖尿病非肾病组131例)和129例健康对照组的热休克蛋白70-2(+1538A/G)基因多态性。结果糖尿病肾病组、糖尿病非肾病组和健康对照组间热休克蛋白70-2(+1538A/G)基因型和等位基因分布均无统计学差异(均P>0.05);与AG基因型组相比,GG基因型组舒张压水平较低,而HDL-C较高(均P<0.05)。二元logistic回归分析进一步显示GG基因型是舒张压增高的保护因素,OR值为0.331(95%CI 0.111~0.983, P<0.05)。结论 HSP70-2(+1538A/G)基因多态性与2型糖尿病肾病的易感性无相关关系,但GG基因型是2型糖尿病患者舒张压增高的保护因素,同时还可能影响HDL-C水平。 Objective To investigate the association between HSP70-2 gene potymorphisms and diabetic nephropathy (DN) in patients with Type 2 diabetes mellitus (T2DM). Methods The study included 130 patients with Diabetic nephropathy (DN), 129 healthy individuals (HC) and 131 patients with T2DM who were free of nephropathy (DM). The genotypes of HSP70-2 +1538 A/G were examined by PCR-restriction fragment length polymorphism (PCR-RFLP) technique. Blood glucose, HbAlc, lipid profiles were also measured. Results There were no statistically differences in genotype distribution and allele frequency of HSP70-2 +1538 A/G among DN, DM and HC groups (P 〉0.05). The diastolic blood pressure was lower in GG genotype group while the high density lipoprotein cholesterol was higher, compared to GA genotype(P〈0.05). Conclusion The polymorphism of HSP70-2 gene 1538A/G is not associated with susceptibility of diabetic nephropathy in Chinese T2DM. The GG genotype was associated with a reduced risk of increased diastolic blood pressure and lower high density lipoprotein cholesterol.
