胃促生长素对大鼠胃排空的影响

Effects of Ghrelin on gastric emptying in rats

目的观察不同剂量的胃促生长素(Ghrelin)腹腔注射后大鼠胃排空效应,阐明Ghrelin在胃排空调节中的作用。方法将50只Wistar大鼠随机分为对照组和4个实验组(A、B、C、D),每组各10只,4个实验组分别采用不同剂量的Ghrelin(0.5、1.0、1.5、2.0 nmol/kg)腹腔注射,对照组采用0.9%氯化钠注射液1ml腹腔注射,45min后每只大鼠半固体糊1.0ml灌胃,30min后处死,测定胃排空率,比较5组间胃排空率的差异。结果腹腔注射Ghrelin后大鼠胃排空率(36.08±5.57)%,与对照组的(27.18±2.37)%相比有明显加快(P<0.05),而且该效应与腹腔注射Ghrelin的量有一定的关系,小剂量Ghrelin(0.5nmol/kg)腹腔注射对胃排空无明显影响(P>0.05),但中、大剂量Ghrelin腹腔注射能明显加速大鼠胃排空效应(均P<0.01)。结论中、大剂量Ghrelin腹腔注射能明显加快健康大鼠的胃排空效应。

Objective To investigate the effects of ghrelin on gastric emptying in rats. Methods Fifty rats were randomly divided into five groups （n=10 in each）. Ghrelin was intraperitoneally injected （0.5, 1.0, 1.5 and 2.0 nmol/kg） in 4 experimental groups; in control group the normal saline was injected instead. Semisolid feed was given by gastric gavage 45 rain after injec- tion, 30min late the animals were sacrificed and gastric emptying was measured. Results The gastric emptying rate in ghrelin groups was significantly higher than that of controls [（36.08 ± 5.57）% vs （27.18 ± 2.37）%] in a dose-dependent manner. Conclu- sion Intraperitoneal injection of ghrelin can accelerate gastric emptying in rats in a dose-dependent manner.
