摘要 采用响应面法优化芡实种皮多酚提取条件,并评价种皮及纯化多酚作为栲胶生产原料的质量。通过单因素试验,确定以丙酮浓度、液料比、超声时间为自变量,以种皮多酚提取含量为响应值,利用Box-Benhnken中心组合方法进行三因素三水平的试验设计,并做响应面分析,建立数学回归模型。按照《中华人民共和国林业行业标准》检验栲胶各指标。结果表明:当丙酮浓度57%、液料比57∶1(mL/g)、超声时间23min时,验证试验的芡实种皮多酚提取含量为113.30mg/g,接近于模型预测值114.00mg/g。同时也证实芡实种皮符合栲胶原料标准,纯化多酚大部分指标与现行其他主要栲胶要求相符,可用于工业生产。 Response surface methodology was applied to optimize the extraction conditions of polyphenols in Semen Euryales seed coat. Obey to analytical test methods of "Forestry industry standards of the People's Republic of China" ,the seed coat and its purified polyphenols quality were evaluated.On the basis of single-factor experiment,the mathematical regression model was established about three independent variables with three levels( acetone concentration, liquid ratio and ultrasonic processing time) and one dependent variable( extraction yield of polyphenols,)through Box- Benhnken center composite design and response surface analysis. The optimum ultrasonic extraction technic was:acetone concentration of 57%, liquid ratio of 57:1 (m!_/g) and ultrasonic time of 23min. Extraction yield of polyphenols was 113.30mg/g by verification testing,which was close to the model predictive value of 114.00mg/g. In addition, tests indicated that seed coat met a criterion of tannin extract raw materials,while its polyphenols was in accordance with the other tannin extract,which could be used for industrial production.
