儿童哮喘患者MBL基因多态性和血清水平的研究

摘要 目的:探讨甘露糖结合凝集素(MBL)基因启动子区单核苷酸多态性(SNP)与哮喘易感性的关系。方法:对61例哮喘患儿和50名正常健康儿童MBL基因启动子区SNP位点-550和-221采用等位基因特异性PCR法(PCR-SSP)检测,并分析其单元型及基因型频率。采用ELISA方法检测儿童哮喘组及儿童健康组血清MBL水平。结果:哮喘组的MBL血清水平比对照组明显增高(P<0.001)。检出HY、LY和LX三种单元型,两组间各单元型比较均无显著性差异。结论:MBL可能参与了哮喘发生发展的过程。 Objective: To investigate the association of single nucleotide polymorphisms(SNP) at positions-550 and-221 in the promoter of the mannose-binding lectin(MBL) gene and the serum protein level of MBL with asthma.Methods: Sequence specific priming-polymerase chain reaction(SSP-PCR) were used to investigate the SNPs of the promoter region of the MBL gene in 61 asthma children and 50 healthy children.Results:The serum protein level of MBL in asthma children was remarkably higher than in con trols(P0.001).Three haplotype,HY,LY,and LX,were present in both groups,and no statistically significant differences were observed between the patients and healthy children.Conclusion:.MBL may be a risk factor for asthma.
