摘要 目的:采用反相高效液相色谱法测定薏苡仁不同饮片炮制品中甘油三油酸酯的含量。方法:采用HPLC-ELSD法,色谱柱为Diamonsil C18色谱柱(250×4.6mm,5μm),流动相为乙腈一二氯甲烷(65 35),流速为1.0 mL min-1,柱温为40℃,采用蒸发光散射检测器,漂移管温度为90℃,氮气流速为1.9 L min-1。结果:甘油三油酸酯在0.1452-2.1780μg范围内,与峰面积呈良好线性关系;平均回收率为96.2%,RSD为1.5%;净薏苡仁、法薏苡仁、麸炒薏苡仁、土炒薏苡仁、清炒薏苡仁的甘油三油酸酯的含量分别为:0.59%、0.90%、0.86%、0.88%、0.91%。结论:本法简便、准确,重复性好,能为江西特色饮片法薏苡仁炮制优势提供科学依据。 Objective:To compare the content of Glycerol Trioleate in the raw medical material and the different processed products of Semen Coicis by HPLC.Method:The Diamonsil C18(250×4.6mm,5μm)was used with mobile phase of acetonitrile-dichloromethanol(65: 35).The mobile phase flow rate was 1.0 mL min-1.The column temperature was controlled at 40℃.The ELSD was used.The tube Temper ature was 90℃,and the flow rate of nitrogen was1.9 L min-1.Result: The calibration curve was linear over the concentration range of 0.1452-2.1780μg for Glycerol Trioleate,The average recovery was 96.2% with RSD=1.5%.The content of Glycerol Trioleate were 0.59% in raw material,0.90%、0.86%、0.88% and 0.91% in different prepared medical material of Semen Coicis respectively.Con clusion: The methods is simple,accurate with a good reproducibility,which can provide scientific basic for the processing advantage of processed medicinal Coix Seed with Jiangxi characteristics.
