摘要
目的:观察冠心微丸(丹参、三七、降香等)对急性心肌缺血模型大鼠心电图和心肌酶的影响。方法:70只SPF级SD大鼠(雌雄各半),随机均分为正常对照组、模型对照组、硝苯地平组(2.7 mg Kg-1,临床等效剂量)、冠心丹参胶囊(0.053 7g生药Kg-1,临床等效剂量)、冠心微丸高剂量(0.107 5g生药Kg-1)、中剂量(0.053 7g生药Kg-1,临床等效剂量)和低剂量(0.026 9g生药Kg-1),预灌胃给药8d,垂体后叶素(Pit)iv.复制急性心肌缺血大鼠模型,于造模5min后十二指肠灌注给药1次,测大鼠心电图、血清心肌酶的含量。结果:Pit注射30s-90s后造模大鼠均出现不同程度的心率减慢(P<0.01)、T波幅值减小、ST段幅值减小或增加、PR与QT间期延长,以及血清肌酶肌酸激酶(CK)(P<0.05)和心乳酸脱氢酶(LDH)(P<0.05)含量异常增加;给药后,各给药组大鼠上述心电图和心肌酶异常均有所改善,硝苯地平组、冠心丹参组、冠心微丸高、中、低剂量组大鼠心率均回复(P<0.01),冠心微丸高、中剂量组血清LDH和CK含量均显著降低(P<0.05)。结论:冠心微丸可改善Pit所致大鼠急性心肌缺血后心电图和心肌酶的异常,其抗急性心肌缺血效应较原剂型有所提高。
Objective:To study the effect of Ganxin Beads on pituitrin-induced acute myocardium ischemia.Methods:After animals were given Ganxin Beads for 8 consecutive days,acute myocardium ischemia animal model was established by femoral venous injection of pituitrin(Pit).Duodenum perfusion therapy of Ganxin Beads was performed 5 min after the model was made successfully.The changes of electrocardiogram(ECG),heart rate and myocardial creatinine kinase(CK) and lactate dehydrogenase(LDH) con tents in serum were examined.Results:After pit was injected for 30s-90s,decreased heart rate、T wave flat or biphasic or inverted、 ST segment elevation or down、PR and QT interval prolongation in ECG were detected in animals of the model.However,Ganxin Beads significantly improved ECG(P0.05) and heart rate(P0.01 or P0.05) and obviously reduced contents of CK and LDH(P0.05).Conclusion:Ganxin Beads could improve ECG and decrease contents of myocardial enzymes in the animal model,which suggested that it may be used in clinical therapy of acute myocardial ischemic diseases.
