冷藏蓝舌病样品的Real-time PCR检测和病毒颗粒稳定性研究 被引量:6

摘要 为了探测长期冷藏对蓝舌病样品的Real-time PCR检测结果和蓝舌病病毒活性的影响,本试验对18份4℃保存了10~18年的样品进行Real-time PCR检测,并对其中10份全血样品进行蓝舌病病毒分离和免疫酶染色,结果显示所有检测样品都为Real-time PCR阳性,从10份全血样品分离出5株蓝舌病病毒,通过免疫酶染色进一步证实为蓝舌病活病毒。研究结果提示冷藏蓝舌病病毒的活性高达18年,其稳定性超过目前现有的认识。 In order to understand the stability of blue-tongue virus (BTV)storage in cold room for long time, 18 tissue sam- ples which were kept in 4 ℃ for 10 to 15 years were test by Real-time PCR, and 10 blood samples which were kept in 4 ℃ for 15 years were subjected to virus isolation process, then was proved by monoclonal antibody staining,research results showed all old samples were positive in Real-time PCR, further more, 5 live virus were isolated from 10 old blood samples. The results suggested the stability of BTV was higher than we thought before.
