摘要
目的:考察玉郎伞多糖(YLSP)对快速老化(SAMP8)小鼠额叶、海马神经细胞半胱天冬蛋白酶-3(Caspase-3)表达及活性的影响。方法:选择6月龄SAMP8小鼠,随机分为模型组,石杉碱甲组(0.02 mg.kg-1),YLSP低、高剂量组(45,180mg.kg-1),选择6月龄正常老化小鼠(SAMR1)作为对照组,以上各组每日上午ig给药l次,连续40 d,于末次给药的次日应用免疫组织化学方法检测额叶和海马Caspase-3的表达变化及神经细胞凋亡情况,测定Caspase-3活性。结果:SAMP8小鼠额叶和海马Caspase-3阳性细胞数分别为(46.00±5.30),(51.36±3.28)个,Caspase-3活性分别为2 380±193,2 473±241,均较对照组显著升高(4.90±0.60),(6.14±0.53);(437±76),(521±79)(P<0.01);与模型组相比,石杉碱甲组、YLSP低、高剂量组额叶和海马Caspase-3阳性细胞数(23.48±3.46,27.86±4.04;24.54±3.55,30.60±2.62;17.98±2.97,23.90±2.58)及Caspase-3活性(1323±126,1 456±130;1 397±133,1 483±147;987±79,1 091±95)明显低于模型组(P<0.01);与石杉碱甲组、YLSP低剂量组比较,YLSP高剂量组的额叶和海马Caspase-3阳性细胞数及Caspase-3活性显著降低(P<0.05)。结论:表明YLSP降低Caspase-3表达及活性,减少SAMP8小鼠脑部神经细胞凋亡。
Objective: The effects of Yulangsan polysaccharides(YLSP) on Caspase-3 expression and activity of the frontal lobe and hippocampus in senescence accelerated-prone mouse 8(SAMP8) mice brain were investigated.Method: The SAMP8 mice were adopted as the Alzheimer's disease model group,and SAMR1 as the control group.Both groups were randomized and given 0.5% CMC-Na solution.Huperzine(0.02 mg.kg-1) was given to drug-positive group and two YLSP groups were given 45,180 mg.kg-1 respectively,continuous administration for 40 days.In the frontal lobe and hippocampus,Caspase-3 expression and activity were detected respectively by immunohistochemical method and Caspase-3 activity assay kit.Result: In model group,Caspase-3 expression and activity were obviously higher than that in the control group(P〈0.01).In drug-positive and YLSP groups,Caspase-3 expression and activity were obviously lower than that in model group(P〈0.01).YLSP presented a dose-dependent manner(P〈0.05).Conclusion: The abnormal Caspase-3 expression and activity can be reduced by YLSP,and it has the protective function towards neural cell apoptosis in SAMP8 mouse brain.
