香港远志黄酮苷对局灶性脑缺血大鼠神经保护及TGF-β、HSP70含量影响 被引量:4

The influence of flavonoids from polygala Hongkongensis on Neuroprotective and the contents of TGF-β and HSP70 in the rats of focal cerebral ischemia

摘要 目的探讨香港远志黄酮苷对局灶性恼缺血大鼠脑神经保护作用和机制。方法线栓法制备SD大鼠大脑中动脉闭塞(MCAO)模型,随机分5组,假手术组、模型组、山柰酚-3-0-葡萄糖苷(PHN-08)干预组、山柰酚-3-0-芸香糖苷(PHN-11)干预组及合方干预组。干预组按设定方式给药,假手术组、模型组给生理盐水。观察MCAO后24h神经功能缺损评分(NBDS)、脑梗死体积(IV)、血清神经元烯醇化酶(NSE)含量,酶联免疫吸附法(ELISA)检测MCAO后12h、24h脑组织匀浆转化生长因子β(TGF-β)和热休克蛋白70(HSP70)含量。结果与假手术组比较,模型组和干预组NBDS、IV、NSE、TGF-β及HSP70显著增高(P<0.05)。与模型组比较,干预组NBDS、IV、NSE显著降低,MCAO后12h,PHN-08组、合方组TGF-β显著增高(P<0.05),MCAO后24h,各干预组TGF-β、HSP70显著增高(P<0.05)。结论香港远志黄酮苷预处理,对局灶性脑缺血大鼠脑神经有保护作用,机制可能是促进TGF-β和HSP70的表达,延缓神经元死亡,防止梗死灶扩大、改善神经行为缺损。 Objective To investigate the neuroprotective effect and Mechanism of flavonoids from polygala hongkongensis on the focal cerebral ischemia in rats. Methods SD rats were randomly divided into 5 groups,sham operated group,model group and treatment groups(kaempferol-3-O-glueoside (PHN-08)group, Kaempferol-3-O-rutinoside (PHN-11)group and combined formuIa group. Treatment groups received drug respectively according to grouping protocol ,the sham operated group and model group received infusions of normal saline. The middle cerebral artery occlusion (MCAO) was made by thread approach, at 24 h after MCAO,the neurological behavior deficits scores(NBDS),infarction volume (IV)and NSE were observed, at 12 h and 24 h after MCAO,the contents of transforming growth factor-β(TGF-β) and beats shock protein70(HSP70) in brain homogenate were detected by enzyme-linked immunosorbent assay (ELISA). Results Compared with sham operated group, the NBDS, IV, NSE, TGF-1;and HSP70 were significantly increased in both the model and treatment groups (P〈0.05). Compared with model group, the NBDS,IV and NSE were significantly decreased in treatment groups,the TGF-β at 12 h after MCAO in PHN-08 and combined formula groups and the TGF-β and HSP70 at 24 h after MCAO in all treatment groups were significantly increased(P〈0.05). Conclusions The flavonoids from polygala hongkongensis given prior to the focal cerebral ischemia in rats play a neuroprotectivc role which may be achieved by increasing the expression of TGF-β and HSP70.
