摘要 目的探讨内脂素(Visfatin)过度表达对多囊卵巢综合征(PCOS)模型大鼠脂代谢的影响。方法采用42 d龄SD清洁雌性大鼠注射脱氢表雄酮(DHEA)和胰岛素诱导建立PCOS模型(n=20),并以等量油剂代替DHEA为对照组(n=20),大鼠Visfatin基因真核表达质粒和空质粒分别转染PCOS组和对照组,ELISA方法测定血浆Visfatin水平;RT-PCR测定Visfatin mRNA表达;TC、HDL-C、LDL-C等在全自动生化分析仪进行测定。结果 Visfatin质粒注射72 h后较注射前,①血浆Visfatin水平显著升高(P<0.05)。②Visfatin mRNA表达显著升高(P<0.05)。③血浆TC、LDL-C水平显著下降;HDL-C水平显著升高(均P<0.05)。结论Visfatin质粒转染在PCOS大鼠血脂中起重要作用。 【Objective】 To study the effects of visfatin gene over-expression on blood lipid level in rats with Polycystic Ovarian Syndrome(pcos).【Methods】 Rat model induced by subcutaneous injecting dehydroeplandrosterone(DHEA) and Insulin on the 42-day-old female SD rats were injected daily with DHEA for up to 28 days and rats in group 2 injected with oil at the same time.The control plasmid pcDNA3.1(+) and recombinant plasmid pcDNA3.1-visfatin were transfected into rats in control group and Polycystic Ovarian Syndrome group.Serum level of visfatin in rats of both groups were determined with ELISA method and the expression of visfatin mRNA by RT-PCR.The blood lipid level was detected with automatic biochemistry analyzer.【Results】 The expression of the serum level of visfatin and visfatin mRNA were significantly increased in after 3 d of pcDNA3.1-visfatin transfection than those in the control plasmid pcDNA3.1(+) rats(P 0.05).The levels of plasma total cholesterol(TC) were significantly decreased,high density lipop rotein cholesterol(HDL-C) were significantly increased and low density lipoprotein cholesterol(LDL-C) were significantly decreased in after 3 d of pcDNA3.1-visfatin transfection than those in the control plasmid pcDNA3.1(+) rats(P 0.05).【Conclusion】 The transfection of visfatin plasmid was related to blood lipid level in rats with PCOS.
