摘要
目的探讨胆管癌发生发展中Notch1、c-Myc及miR145表达的相互关系。方法建立胆管癌小鼠模型并检测胆管癌不同发展阶段中Notch1、c-Myc及miR145的表达。通过慢病毒转染胆管癌QBC939细胞检测Notch1、c-Myc及miR145三者之间表达的相互关系。结果成功建立胆管癌小鼠模型,检测不同分化程度的胆管癌小鼠肝组织中Notch1、c-Myc及miR145的表达发现,Notch1及c-Myc的表达与胆管癌发展呈正相关,而miR145呈负相关。Notch1 siRNA慢病毒转染胆管癌QBC939细胞可见c-Myc表达随Notch1表达降低而降低,miR145表达则随Notch1表达降低而升高。同时发现,载有过表达miR145的慢病毒转染胆管癌QBC939细胞后,c-Myc的表达下调。结论 Notch1/miR145/c-Myc通路可能与胆管癌的发生发展有关。
【Objective】 To study on the expression relationship between Notch1,c-Myc and miR145 in human cholangiocarcinoma.【Methods】 Establish a mice model of CCA and detect the expression of Notch1,c-Myc and miR145 in CCA progression.Test the expression relationship between Notch1,c-Myc and miR145 though lentiviral transfection.【Results】 We built a successful mice model of CCA,and found that Notch1 and c-Myc expression have positive correlation with CCA progression,whereas the expression of miR145 has negative correlation with CCA progression.The expression of c-Myc and miR145 in QBC393 cells infected with lentiviruses driving expression of shRNA targeting Notch1 were down-regulated and up-regulated separately.The expression of c-Myc was down-regulated in QBC939 cells infected with lentiviruses driving overexpressed miR145.【Conclusion】 The Notch1/c-Myc/miR145 pathway has the relationship with the progression of human cholangiocarcinoma.
