宫颈上皮内瘤变中不同高危型HPV感染与p53表达的相关性研究

Study on correlation between different high risk human papillomavirus(HPV) groups and expression of p53 in cervical intraepithelial neoplasia cases

目的探讨宫颈上皮内瘤变(CIN)病例中不同高危型人乳头瘤病毒(HPV)感染时p53的表达情况。方法 60例CIN病例(HC2检测高危型HPV为阳性,宫颈活检组织学证实为CIN)的石蜡标本,采用原位杂交方法检测HPV16/18、HPV31/33,并用免疫组织化学检测其p53蛋白的表达。以20例宫颈鳞状细胞癌为阳性对照,20例正常宫颈为阴性对照。结果 CIN病例中HPV16/18感染组、HPV31/33感染组及其他高危型HPV感染组(HPV35、39、45、51、52、56、58、59、68)p53阳性表达率分别为15.56%、13.75%、17.14%,三者之间差异无显著性(P<0.05)。结论不同亚型高危型HPV感染的CIN病例,p53表达率相近,推测不同亚型高危型HPV对宿主p53影响可能相同,与宿主p53结合的量可能相同;CIN与不同高危型HPV的相关程度和p53的突变可能无明显相关性。

【Objective】 To compare the expression of p53 in different high risk human papillomavirus（HPV） types in cervical intraepithelial neoplasia（CIN） cases.【Methods】 60 paraffinembedded samples（infected with high risk HPV with method of hybrid capture Ⅱ,HC2,and diagnosised with CIN in histology） were used to detect HPV16/18 and HPV31/33 with method of in situ hybridization,and to detect the expression of p53 protein with method of immunohistochemistry.20 cervical squamous cell carcinoma cases and 20 normal cervical cases were served as positive group and negative group respectively.【Results】 The positive expression rates of p53 protein were 15.56%,13.75%,17.14% respectively in CIN cases infected with HPV16/18,HPV31/33 and other high risk HPV group（HPV35,39,45,51,52,56,58,59,68）,without statistic difference（P 0.05）.【Conclusion】 Expression rate of p53 was similar in CIN cases infected with different high risk HPVs.It suggest high risk HPVs may have the same influence upon the host＇s p53,different high risk HPVs may have the same binding form with host＇s p53,and the relationship between CIN degree and there may be no correlation between different high risk HPVs and p53 mutation.
