摘要
以朱槿曲叶病毒病株为材料,结合总DNA提取的CTAB法和质粒提取的离心柱方法,发展一种简单快速的多糖植物双生病毒基因组核酸的提取方法,用聚合酶链式反应(PCR)和实时定量PCR予以验证。结果显示:与目前的2种常用方法相比,改进的方法在灵敏度、时间、价格和产率上都具有一定的优势,更适合于双生病毒基因组核酸的提取和检测应用。
The geminous viruses are a group of single-stranded circular DNA viruses occurred worldwide, and have caused severe diseases in several economically important crops. With the material of Hibiscus rosa-sinesis caused by Cotton leaf curl Multan virus, a simple and rapid method for extraction of genome from plant rich in polysaccharide was developed by combining the total DNA extracted CTAB method and the plasmid extraction method of centrifugal column. The results show that the new modified method has more advantages on sensitivity, experiment length, price and production than other two methods and is more suitable for the extraction and detection of genome from plant rich in polysaccharide.
