乳化异氟醚预处理对大鼠内毒素所致急性肺损伤的作用 被引量:2

Effects of Emulsified Isoflurane Preconditioning on LPS-induced Acute Lung Injury in Rats

摘要 目的研究乳化异氟醚(EISO)预处理对大鼠内毒素(LPS)所致急性肺损伤是否具有保护作用。方法健康雄性SD大鼠,分为急性肺损伤模型组(LPS组,n=10)、乳化异氟醚组(EISO组,n=10)、脂肪乳组(INT组,n=10)和对照组(CON组,n=5),按分组分别静脉泵注2mL生理盐水、EISO 4mL/(kg.h)、脂肪乳(INT)、生理盐水2mL预处理30min,然后LPS组、EISO组和INT组静脉注入LPS 8mg/kg,CON组静脉注入生理盐水。监测动脉血压和心率。5h后,测动脉血气分析;测量血浆肿瘤坏死因子-α(TNF-α),白介素-6(IL-6)水平;测量肺湿干重比值(W/D);测量肺组织髓过氧化物酶(MPO)、丙二醛(MDA)、超氧化物歧化酶(SOD)水平。结果与CON组相比,其余3组氧合指数降低,肺W/D增加,血浆TNF-α、IL-6浓度升高,肺组织MPO活性增加,MDA含量增加,SOD活性降低(P<0.05);与LPS组相比,EISO组氧合指数升高,肺W/D降低,血浆TNF-α、IL-6浓度降低,肺组织MPO活性降低,MDA含量减少,SOD活性增加(P<0.05)。结论乳化异氟醚预处理可以减轻大鼠LPS所致急性肺损伤。 Objective To investigate the effects of emulsified isoflurane preconditioning on LPS-induced acute lung injury in rats. Methods Acute lung injury was induced by the administration of LPS 8 mg/kg intravenously in male Sprague-Dawley rats. The ratswere randomly assigned to 4 groups: LPS-induced acute lung injury group (LPS, n= 10) ; emulsified Isoflurane Preconditioning group (EISO, n= 10) ; Intralipid pre-treated group (INT, n= 10), and saline pre-reated control group (CON, n= 5). In the later 3 groups, 2 mL/kg NS, IL and EISO were infused intravenously for 30 min before the administration of LPS respectively. The animals were then observed for 5 hours. The mean arterial blood pressure(MAP) and heart rate(FIR) were monitored hourly. Arterial blood gas (ABG) was measured for oxygenation index(Ol) at the end of 5h. The blood samples were taken for the measurements of plasma tumor necrosis factor-α(TNF-α) and interleukin-6(IL-6). The lungs were removed for the measurements of wet/dry (W/D) weight ratio, as well as the levels of malondialdehyde (MDA), superoxide dismutase(SOD) ,myeloperoxidase(MPO). Results The OI of group CON was 435±30,group LPS 225±9,group EISO 367±41 ,and group INT 267=k41. Compared with group CON, OI and pulmonary SOD activity significantly decreased while plasma TNF-α,IL-6 concentration and pulmonary MDA level, MPO activity increased in LPS, EISO and INT groups. The plasma TNF-α, IL-6 concentration, pulmonary MDA level and pulmonary MPO activity of group EISO were lower while OI and pulmonary SOl) activity were higher than those of group LPS. Conclusion The emulsified isoflurane pretreatment can ameliorate LPS-induced acute lung injury in rats.
