摘要
载脂蛋白极低密度脂蛋白Ⅱ(ApoVLDL-Ⅱ)是极低密度脂蛋白的主要成分,在禽类繁殖和脂质转运中起着重要作用。试验采用PCR-SSCP法与PCR产物直接测序技术相结合对天柱番鸭ApoVLDL-Ⅱ基因全编码区进行多态性检测,并分析其对肉质和血清生化指标的遗传效应。结果表明,在ApoVLDL-Ⅱ基因外显子3检测到T1986C同义突变,产生CC,CD和DD 3种基因型,等位基因C和D;在3'-UTR检测到C2901T突变,产生AA,AB和BB 3种基因型,等位基因A和B;合并基因型显示,产生3种聚合基因型CC/AA,CD/AB和DD/BB,基因型CC/AA和等位基因C/A的频率分别为0.716 1和0.851 9。最小二乘分析显示,CC/AA基因型个体的血清总蛋白和球蛋白显著高于CD/AB型个体(P<0.05),白球比值和免疫球蛋白A显著低于CD/AB型个体(P<0.05)。研究结果揭示,T1986C和C2901T位点对血清中蛋白含量可能有一定的相关性。
Very low density apolipoprotein-II(ApoVLDL-II) is a major constituent of very low density lipoprotein and is involved in reproduction and lipid transportation in avian species.The polymorphisms of Tianzhu Muscovy ApoVLDL-II gene whole CDS region were detected by using the methods of PCR-SSCP with DNA sequencing,and its relationship with meat quality and biochemical levels were analyzed.The results indicated that the synonymous mutation T1986 C was found in ApoVLDL-II gene exon 3,which resulted in three genotypes CC,CD and DD,and allele C and D.The C2901 T mutation was detected in 3 '-UTR,and produced genotypes AA,AB and BB,and allele A and B.Three polymn genotypes CC / AA,CD / AB and DD / BB were found by two SNPs in-teraction.Genotype CC / AA and allele C / A frequencies were 0.716 1 and 0.851 9,respectively.CC / AA genotype had higher total protein(TP) and globulin(GLO) than CD / AB genotype(P〈0.05).For Alb/GLO ratio and IgA,genotype CC/AA had lower than genotype CD/AB(P〈0.05).The results suggested that the T1986C and C2901T loci of ApoVLDL-II gene may be linked with some serum protein content.
