摘要
以云南含笑冬芽、春芽及新叶为外植体,接种于含有不同激素配比的MS培养基中,诱导愈伤组织及不定芽的形成,比较HgCl2消毒的最佳时间及活性炭、AgNO3抗褐化效果。结果表明:外植体以冬芽为佳,HgCl2最佳消毒时间为18 min;活性炭和AgNO3均有抗褐化的效果,以3.0 mg/L AgNO3抗褐化的效果最为理想;继代增殖效果最好的培养基为:MS+2.0 mg/L 6-BA+0.1 mg/L NAA,平均每株增殖数为3.2;壮苗效果最好的培养基为:MS+1.0 mg/L 6-BA+0.3 mg/L NAA,平均每苗高为3.5 cm;云南含笑春芽、新叶都能诱导出愈伤组织,其中春芽诱导愈伤组织的能力明显高于新叶,诱导率达46.67%。
The winter buds,spring buds and new leaves of Michelia yunnanensis were used as explants,and inoculated in MS media supplemented with different concentrations of 6-BA and NAA to induce adventitious buds and callus.The effects of 0.1% HgCl2 on disinfecting the explants,and the effects of activated carbon and AgNO3 on overcoming browning were studied.The results showed that winter buds were the best explants and 18 min was the best time of 0.1% HgCl2 treatment.Both activated carbon and AgNO3 could overcome M.yunnanensis browning,and 3.0 mg / L AgNO3 worked best.MS + 2.0 mg / L 6-BA + 0.1 mg / L NAA was the best medium for subculture,and the propagation ratio was 3.2;MS + 1.0 mg / L 6-BA + 0.3 mg / L NAA was the best medium for strength the shoots,the average height of shoots was 3.5 cm.Both spring buds and new leaves could induce callus.The induction rate of the spring buds was higher than the new leaves significantly,and its 46.67%.
