摘要
以杨树叶片为试验材料,采用改良SDS法提取基因组DNA,分析5种不同保存方法对样品提取效果的影响。通过A260/A280、琼脂糖凝胶电泳、ISSR-PCR扩增分析对提取的基因组DNA进行鉴定,结果表明,低温保存杨树叶片是比较理想的方法,而在远距离野外资源调查收集时,采用硅胶干燥保存样品是比较可行的。
Effects of different preservation methods on the extraction of genomic DNA with improved SDS method from poplar leaves were studied.The extracted DNA was tested by using the value of A260 /A280,the agarose gelelectrophoresis,and ISSR-PCR amplification.The results showed that low temperature preservation of poplar leaves was the best method.However,silica gel preservation was a feasible alternative for preserving the samples collected the field resources investigation.
