毛葡萄茎尖的离体培养与植株再生 被引量:3

摘要 为了建立毛葡萄茎尖离体培养及植株再生体系,给野生毛葡萄种质资源的超低温保存和无毒苗木培育提供参考,以花溪-4和农院-11毛葡萄组培苗的茎尖为材料,研究植物生长调节剂及浓度配比对毛葡萄茎尖离体培养与植株再生的影响。结果表明:毛葡萄茎尖离体培养与增殖培养的适宜培养基为MS+BA 1.5 mg/L+IBA 0.02 mg/L,离体培养成活率达93.8%以上,增殖率和增殖系数分别为88.6%和5.8;茎尖生根培养的适宜培养基为1/2MS+IBA 0.05~0.15 mg/L+IAA 0.2 mg/L,生根成苗率为100%,建立了毛葡萄茎尖组织培养和植株再生的技术体系。 Effects of proportion and concentration of plant growth regulators on shoot-tip culture and regeneration in vitro was studied using two individuals(Vitis heyneana Huaxi-4 and Nongyuan-11 as materials to construct the in vitro shoot-tip culture and regeneration system of V.heyneana.The results showed that MS+6-BA 1.5 mg/L+IBA 0.02 mg/L was an appropriate medium for shoot-tip culture and regeneration of V.heyneana.The survival rate and reproducibility rate were more than 93.8 % and 88.6 %,respectively.Multiplication coefficient was more than 5.8.1/2MS+IBA 0.05~0.15 mg/L+IAA 0.2 mg/L was an optimal medium for rooting culture,the rooting rate was 100 %.The system for in vitro shoot-tip culture and regeneration of V.heyneana was successfully established.
